针刺人中穴对MCAO大鼠中枢神经系统神经干细胞增殖影响的研究

摘    要:目的]探讨针刺人中穴治疗缺血性脑卒中的神经发生机制。方法]用BrdU+/nestin+免疫荧光双标和蛋白免疫印迹法观察针刺对线栓法大鼠大脑中动脉阻断实验模型(MCAO)大鼠大脑新生神经细胞及皮质、海马、纹状体nestin蛋白表达量的变化的影响。结果]针刺组MCAO大鼠BdrU+、nestin+和BrdU+/nestin+细胞数均高于正常组、假手术组和模型组(P0.05),模型组高于正常组和假手术组(P0.05);针刺组nestin在海马和皮质的表达量高于其他组别(P0.05)。结论]缺血缺氧能引起MCAO大鼠脑神经干细胞的增殖,而针刺人中穴能增强神经干细胞增殖作用,促进大脑功能的修复。

Abstract:Objective] To discuss the mechanism of treating ischemic stroke by acupuncturing Renzhong. Methods] We observed the population of BrdU+/nestin+ cells and expression of nestin protein in cortex, hippocampal and striatum in MCAO rat by double-labeling immunofluorescence and Western blot. Results] Renzhong group's population of BdrU+, nestin+ and BrdU+/nestin+ cells were significant higher than that in normal group, sham group and control group in brain of MCAO rat(P<0.05). Renzhong group also had the highest ex-pression of nestin protein in cortex and hippocampal(P<0.05). Conclusion] Acupuncture of Renzhong can enhance the proliferation of NSCs in central nervous system of MCAO rat.
