光动力作用对人乳腺癌细胞增殖的实验研究

摘    要:目的:探讨光动力疗法(PDT)对人乳腺癌细胞增殖的影响。方法:以人乳腺癌细胞株MDA-MB-231为研究对象,以血卟啉衍生物(HpD)为光敏剂,630nm波长的激光为光源,MTT法绘制MDA-MB-231PDT后不同时间的生长曲线。流式细胞仪检测PDT对人乳腺癌细胞阻滞的细胞周期,免疫组化观察PDT对增殖细胞核抗原(PCNA)的影响。结果:生长曲线示PDT对人乳腺癌细胞增殖有明显的抑制作用;PDT作用后早期(12h)G0/G1期比例明显高于对照组(P<0.05),且呈时间依赖性变化;免疫组化示:实验组PDT后各时间段PCNA阳性细胞蛋白表达率较对照组阳性表达率明显下降(P<0.01)。结论:光动力疗法可阻滞人乳腺癌细胞增殖,其机制可能与PDT阻滞细胞于G0/G1期和降低PCNA的表达有关。

Abstract:Objective:To observe the proliferation effect of photodynamic therapy(PDT) on human breast cancer cell line MDA-MB-231 in vitro.Methods:Human breast cancer cell line MDA-MB-231 was used to investigate the killing role of PDT with HpD as photosenitizer and 630nm successive laser radiation.The growth curve was described according to MTT colorometric assay;Cell cycle blockage effect of breast cancer cell line MDA-MB-231 in PDT was observed by flow cytometry;PCNA was detected in PDT by immunocytochemistry.Results:PDT could obviously inhibit the growth of breast cancer;G0/G1 phase cell cycle blockage was induced after 12 hours of HpD-PDT(P<0.05),and the percentage of G0/G1 phase was significantly raised along with the time prolongation of HpD-PDT;The percent of PCNA was lower than control group(P<0.01).Conclusion:Cell proliferation in human breast cancer MDA-MB-231 was obviously inhibited by HpD-PDT,the mechanism may be related to G0/G1 phase cell cycle blockage and the decreased PCNA.
