| 摘 要： | 本文报道用hCG免疫的BALB/c小鼠脾细胞,与小鼠骨髓瘤细胞NS-1融合,获得6株分泌抗hCG单克隆抗体的杂交瘤细胞。其中,2B5和2D8二株细胞的分泌抗体,对hCG-β亚基呈强阳性反应。其培养液抗体的ELISA效价为10~(-2)～10~(-4);注入同系小鼠腹腔诱生的腹水抗体效价达10~(-5)～10~(-7)。2B5抗体为IgA,2D8抗体属IgG_1。它们对hCG-β的亲和常数(K_a)分别为0.8×10~9升/克分子和1.8×10~9升/克分子.在以~(125)I-hCG-β作为标记物的检测系统中,2B5抗体在50％抑制时同hLH的交叉反应为5.7％,而2D8抗体为3.1％.

| Abstract: | Six hybridoma cells have been developed by fusion of NS-I myeloma cells with spleno-cytes of BALB/c mice immunized with hCG. Two of these cell lines, referred to as 2B5 and 2D8, could secrete the monoclonal antibodies specific for hCG-β in the culture medium, Antibody titers of the culture by ELISA were 10-2-10-4, while that of the ascites fluid were 10-5-10-7. The antibodies of 2B5 were of IgA and that of 2D8 were of IgG1. The cross-reactivity of the antibodies of 2B5 and 2D8 with hLH at 50 per cent inhibition was 5.7% and 3.1% respectively. The affinity constant (Ka) of the antibodies 2B5 was 0.8×10(?)L/M and that of 2D8 was 1.8×109/M. They have been used in clinical diagnosis, tumour imaging and affinity chromatography, and the results will be published. |
