Abstract:Objective To explore the mechanism of lncRNA KCNQ1OT1 on ischemia-reperfusion cardiomyocyte apoptosis.Methods H9c2 cells were used to prepare the Hypoxia/reoxygenation(H/R)model,and the expression of cardiac myocytes KCNQ1OT1 and miR-760 were regulated by transfection,and randomly divided into control group,model group,negative control group,si KCNQ1OT1 group and miRNA inhibitor group.CCK-8 was used to detect the effect of KCNQ1OT1 on cardiomyocyte viability,flow cytometry was used to detect the effect of KCNQ1OT1 on cardiomyocyte apoptosis,and western blot was used to detect the effect of KCNQ1OT1 on the expression of cardiomyocyte apoptosis-related proteins Bcl-2,Bax and cleaved-Caspase-3.Results The cardiomyocyte viability of the model group was significantly lower than that of the control group(P<0.01),the cardiomyocyte viability of the si KCNQ1OT1 group was significantly higher than that of the model group and the miRNA inhibitor group(P<0.01).The apoptosis rate of cardiomyocytes in the model group was significantly higher than that of the control group(P<0.01),the apoptosis rate of cardiomyocytes in the si KCNQ1OT1 group was significantly lower than that of the model group and the miRNA inhibitor group(P<0.01).The expression of Bcl-2 in cardiomyocytes of the model group was significantly lower than that of the control group(P<0.01),and the expression of Bax and cleaved-Caspase-3 was significantly higher than that of the control group(P<0.01).The expression of Bcl-2 in cardiomyocytes of si KCNQ1OT1 group was significantly higher than that of model group and miRNA inhibitor group(P<0.01),and the expression of Bax and cleaved-Caspase-3 were significantly lower than those of model group and miRNA inhibitor group(P<0.01).Conclusions KCNQ1OT1 regulates apoptosis of myocardial cells induced by ischemia reperfusion by down-regulating the expression of miR-760,which is related to the regulation of the expression of bcl-2,Bax and cleaved caspase-3 proteins.
