肝脏中肾综合征出血热病毒感染与c-myc基因的表达以及免疫球蛋白的异常分布

摘    要:目的:观察比较人和动物肝脏感染肾综合征出血热(HFRS)病毒后病变的异同.方法:以原位分子杂交和免疫组化检测HFRS尸检及自然感染大鼠肝脏中病毒RNA,病毒抗原及免疫球蛋白(Ig)的定位和分布及尸检肝脏中c-myc基因.结果:尸检肝脏中病毒RNA,病毒抗原和Ig的阳性率分别为84.21%(16/19),88.23%(15/17)和53.30%(8/14),大鼠肝脏阳性率分别为92.90%(79/85),93.10%(27/29)和54.50%(18/29);尸检肝脏c-myc基因阳性率为50.00%(8/16);尸检阳性者主要为死于休克期和重型病例;类似于人尸检肝脏中的病变未在大鼠肝脏中观察到.结论:与其它脏器相比,肝脏具有较高的病毒感染率.肝细胞异常Ig阳性可能是由病毒感染造成的肝细胞的可逆性损伤的体现,而肝细胞的坏死可能是损伤严重者.肝脏局部血液循环障碍是造成大片坏死的主要因素.C-myc基因的表达可能是肝脏抗损伤的一种反应.

Hemorrhagic fever with renal syndrome virus infection in livers withc-myc oncogene expression and immunoglobulin abnormal distribu-tion
Abstract:Objective: Comparative study of autopsy and naturally infected rat livers was carried out to getmore information about infection and its correlation with alternation in livers. Methods: In situ hybridizationwas used for detection of viral RNA and c-myc oncogene and immunocytochemical method was used for detec-tion of viral antigen and immunoglobulin(Ig).Results:The positive rates of viral RNA,antigens, Ig and c-myc in autopsy livers were 84.21%(16/19),88.23%(15/17),53.30%(8/14)and 50.00%(8/16)respec-tively,and detected mainly in the patients died in the early phases of the disease. Similar distribution and lo-calization of viral RNA,antigens and Ig were also found in the infected rat livers and their positive rates re-spectively 92. 90%(79/85 ), 93. 10%(27/29)and 54. 50%(18/29)respectively.No similar pathologicalchanges as in autopsy livers were seen in rat livers. Couclusion: Livers conceived higher positive rates for vi-ral RNA and antigen comparing to the other tissues in the body. Abnormal localization of Ig in hepatocytesmight be a demonstration of reversible damage caused by virus infection, and necrosis might be one worse endof the lesion. The local infarct-like necrosis of hepatocytes could possibly be caused by poor local blood sup-ply. The c-myc oncogene expression in livers may be a response of the tissues to the damage.
