胰腺癌细胞ASPC-1凋亡过程中caspase 1和2的表达

目的 :探讨细胞凋亡过程中 caspase- 1和 caspase- 2的表达及其意义。　方法 :胰腺癌细胞系 ASPC- 1无血清培养 2 4h后加入 IFN- γ,于 6、1 2和 2 4h收集细胞 ,检测细胞凋亡情况 (流式细胞法 )和 caspase- 1和 caspase- 2蛋白质表达 (western blot法 ) ,以不加 IFN -γ的细胞为对照组。　结果 :IFN-γ刺激可诱导 ASPC- 1细胞的凋亡 ,随着时间延长凋亡数量增多 ,2 4h达 9%。Caspase- 1和 caspase- 2表达随时间而增强 ,对照组轻度表达。　结论 :Caspase- 1和 cas-pase- 2在细胞凋亡过程中表达 ,是重要的凋亡调节基因

Role of caspase-1 and 2 in apoptosis of ASPC-1 cells

ObjectivesTo evaluate the expression of caspase-1 and caspase-2 in apoptosis.　 Methods ASPC-1 cells (a pancreatic cancer cell line) were cultured in serum-free DMEM medium for 24 hours. IFN-γ was added to the medium, and cells were collected at 6, 12 and 24 hours. Apoptosis was measured by flow cytometry. Caspase-1 and caspase-2 expression was measured by western blot analysis. Cells without IFN-γ were used as controls. 　Results Stimulation with IFN-γ induced apoptosis of ASPC-1 cells, the number of apoptotic cells increased to 9% at 24 hours. Both caspase-1 and caspase-2 were expressed in the IFN-γ stimulated cells and strengthened with time. Caspase-1 and caspase-2 was slightly expressed in the control group.　 Conclusions; Caspase-1 and caspase-2 were expressed in ASPC-1 cell apoptosis, indicating a role of caspases in apoptosis.
