抗CD28抗体协同刺激增强抗CD3抗体体外激活T淋巴细胞并降低TGF-β的表达

摘    要:为了探讨体外激活T淋巴细胞的方法及活化后T淋巴细胞的免疫学特征,利用抗CD3、抗CD28抗体在体外刺激健康人外周血单个核细胞PBMNC,检测淋巴细胞转化,CD8^+CD25^+细胞比例和肿瘤生长因子β(TGF-β)表达等相关免疫学指标.结果表明,在抗CD28抗体的协同作用下,抗CD3抗体的刺激活性明显提高,CD8^+CD25^+细胞比例明显增加,TGF-β的表达下调.结论:抗CD28抗体协同刺激可以提供第二信号,使T淋巴细胞充分活化,TGF-β表达水平明显下降.

Abstract:In order to study how to activate T cells and their immunological characteristics, the anti-CD3 and anti-CD28 McAbs were used to stimulate PBMNC, then their related immunological changes, such as lymphocyte transformation function, the percentage of CD8(+)CD25(+) cells and TGF-beta expression were deleted by lymphocyte transformation assay, flow cytometry and RT-PCR respectively. The results showed that in costimulation with anti-CD28 antibody stimulation, the activity of anti-CD3 antibody was significantly enhanced, the ratio of CD8(+)CD25(+) cells of T cells was obviously increased, while TGF-beta expression was down-regulated. It was concluded that the anti-CD28 antibody costimulation could provide stimulatory signal II, which make T cells more active, while the expression of TGF-beta significantly down-regulated.
