CDC25A、Smad3在食管鳞癌中的表达及其相互关系

摘    要:目的探讨CDC25A、Smad3在食管鳞癌组织中表达的意义及CDC25A蛋白与Smad3蛋白、细胞增殖指数、细胞凋亡率的关系。方法采用免疫组织化学S-P法检测CDC25A蛋白在52例食管鳞状细胞癌、24例正常黏膜中的表达。用流式细胞术检测其中30例食管鳞癌、5例正常黏膜的细胞凋亡率、增殖指数及CDC25A蛋白、Smad3蛋白的表达。结果CDC25A在癌组织中的阳性表达率为50%,明显高于正常黏膜(P〈0.05),CDC25A在高分化和中低分化组、浸润至纤维膜和未浸润至纤维膜组、有淋巴结转移和无淋巴结转移组中的差异显著(P〈0.05)。Smad3蛋白在癌组织中的表达低于正常黏膜(P〈0.05)。Smad3蛋白与CDC25A蛋白的表达呈负相关(r=-0.482,P=0.007)。核膜CDC25A蛋白阳性者的增殖指数高于核膜CDC25A蛋白阴性者(P〈0.05);细胞质CDC25A蛋白表达阳性者的凋亡率低于细胞质CDC25A蛋白表达阴性者,但差异无统计学意义(P〉0.05)。结论Smad3蛋白的下调可能与食管癌中CDC25A蛋白含量升高有关;细胞核中的CDC25A蛋白可能具有促增殖作用;CDC25A蛋白可能参与了食管癌的发生、发展。

Abstract:Objective To investigate the expressions of CDC25A and Smad3 in esophageal squamous cell carcinoma and the relationship of CDC25A with Smad3 protein,cell apoptosis,proliferation index(PI).Methods The expression of CDC25A was examined by SP immunohistochemical stain(IHC)in 52 esophageal carcinoma specimens and 24 normal tissues.Moreover,CDC25A protein,Smad3 protein,cell apoptosis,proliferation index(PI)were examined by FCM in 30 esophageal carcinoma specimens and 5 normal tissues.Results The positive expression rate of CDC25A in esophageal carcinoma was 50%,significantly higher than 0% in normal tissues(P<0.05).High expression of CDC25A was correlated with invasion,tumor differentiation and metastasis of esophageal squamous cell carcinoma(P<0.05).The positive expression of Smad3 protein in esophageal carcinoma was significantly lower than that in normal tissues(P<0.05).CDC25A protein expression was negatively associated with Smad3 protein expression(P<0.05).The apoptosis rate in the cells with the positive expression of CDC25A protein in cytoplasm was higher than that of negative CDC25A expression,but without statistical significance.PI of the cells with the positive expression of CDC25A protein in nuclear membrane was higher than that of negative expression(P<0.05).Conclusion The reduction of Smad3 protein was correlated with the abundance of CDC25A protein.The CDC25A protein in nuclear membrane might contribute to cell proliferation.CDC25A protein may be involved in the carcinogenesis and progression of esophageal squamous cell carcinoma.
