山楂的HPLC指纹图谱研究

摘    要:目的:采用高效液相色谱法(high performance liquid chromatography,HPLC)建立山楂药材指纹图谱,以控制其质量。方法:以绿原酸为参照物,采用HPLC分析山楂内有效成分,并利用计算机相似性评价系统对指纹图谱进行相似度分析。色谱柱为Diamonsil~C18(250mm×4.6mm,5μm),采用冰乙酸和乙腈组成的缓冲溶液梯度洗脱,流速0.8ml/min;检测波长325nm。结果:精密度和重复性试验中,相对峰面积大于10%的共有峰的RSD均小于5%,符合《中药注射剂色谱指纹图谱实验研究技术指南》相关要求。结论:本研究建立的方法简单,重复性良好,可作为控制山楂药材的内在质量的标准。

Abstract:Objective: To establish the fingerprint chromatography of crude Hawthorn berry using high performance liquid chromatography (HPLC) for quality control. Methods: Caffeotannic acid was used as internal standard compound and the active components of crude Hawthorn berry was analyzed by HPLC. Computer aided similarity evaluation was used for analyzing the similarity. Diamonsil-C18 (250 mm×4.6 mm,5 μm)was used, the mobile phase was consisted of glacial acetic acid and acetonitrile with gradient elution at flow rate of 0.8 ml/min, and the detection wavelength was 325 nm. Results: In precision test and reproducibility test, the RSD of common peak areas with relative peak areas more than 10% of total peak areas was less than 5%, which accorded with related requests of Experimental Technology Guides for Fingerprint Chromatography of Traditional Chinese Medicine Injection. Conclusion: The methods established in this research are simple and reproducible, and it can be used for the quality control of crude Hawthorn berry.
