摘    要:目的:研究神经干细胞(NSCs)移植对大鼠脊髓损伤(SCI)后轴突再生及后肢运动功能的影响,为NSCs移植治疗SCI提供实验依据。方法:5-溴脱氧尿嘧啶核苷(BrdU)法标记处于对数生长期的NSCs,采用电控SCI打击装置制作大鼠SCI模型。设立NSCs移植组、SCI组、假手术组各10只。NSCs移植组大鼠SCI后3d进行NSCs移植。应用免疫组织化学染色观察BrdU标记的移植细胞的存活、迁移情况以及NR200的表达,采用行为学(BBB)评分法观察大鼠后肢运动功能的恢复情况。结果:NSCs移植组在移植区域及其邻近区域可检测到BrdU阳性NSCs;NF200免疫阳性细胞平均光密度值较SCI组明显增高(P〈0.05);NSCs移植组术后第28天NF200免疫阳性轴突数目较SCI组明显增多(P〈O.01);BBB评分亦明显高于SCI组(均P〈0.05)。结论:体外培养的胚胎大鼠NSCs在移植到SCI区域后可存活、迁移,参与了SCI处神经轴突的结构重建,可促进大鼠后肢运动功能恢复。
