PMA诱导节律基因表达对细胞紫外线损伤的影响

摘    要:目的研究诱导NIH3T3细胞节律基因表达对细胞紫外线损伤的影响。方法用体外节律诱导剂乙酰肉豆蔻佛波酯诱导NIH-3T3细胞节律基因表达。分别选择mPer2基因表达的谷值和峰值时刻对细胞进行紫外线照射,以非诱导节律的单纯照射组为对照,通过流式细胞术检测细胞凋亡和细胞周期分布,平板克隆形成试验检测细胞增殖情况。结果mPer2基因表达峰值时进行紫外线照射的细胞,与mPer2基因表达谷值时和非诱导节律紫外线照射细胞比较,增殖能力强、凋亡率低、G1期阻滞明显。结论节律基因诱导的NIH3T3细胞的紫外线损伤减小,并与mPer2的基因表达相关,其保护作用可能与mPer2基因过表达有关。

Abstract:Objective To study the effects of circadian gene expression induced by rhythm inductor on NIH3T3 cells injury caused by ultraviolet irradiation.Methods Phorbol 12-myristate 13-acetate(PMA) was used to induce the circadian genes expression.Ultraviolet irradiation was given on cells at two different time points when the mPer2 expression was at peak and at trough. After the irradiation,the cell apoptosis and distribution in cell cycle were detected by flow cytometry,and colon formation assay was made to detect the cell proliferation.Results Compared with the cells without induction by PMA and the cells of mPer2 expression at trough,the cells of mPer2 expression at peak demonstrated lower rates of apoptosis,higher percentage of cells at G1 stage and higher colon forming efficiency.Conclusion After Ultraviolet irradiation,circadian gene expression induced by PMA may diminish irradiation injury of cells,and this protection may be related to mPer2 expression.
