XRCC1基因多态性与铅中毒易感性关系的研究

摘    要:目的:探讨XRCC1基因多态性与铅中毒易感性的关系。方法:采用病例-对照研究,收集南通市某钢丝厂137例铅作业工人,其中61例确诊铅中毒患者作为病例组,其余76例铅接触工人作为内对照,选取正常人群75例作为外对照。应用聚合酶链反应-限制性片段长度多态性技术(PCR-RFLP)方法分析3组人群XRCC1A194W和A399Q基因型频率,比较不同基因型与铅中毒易感性的关系。结果:XRCC1 A194W的CT+TT在病例组和内对照组的分布频率差异有统计学意义(P<0.05),CT+TT基因型能增加铅中毒的易感性(OR=2.46,95%CI=1.16~5.21);XRCC1 A399Q的GA+AA在病例组和外对照组的分布频率差异有统计学意义(P<0.05),携带GA+AA基因型的个体发生铅中毒的危险性降低(OR=0.31,95%CI=0.15~0.65)。结论:XRCC1基因A194W和A399Q的多态性可能与铅中毒易感性有关。

Abstract:Objective:To explore the relationship between gene polymorphism of XRCC1 and susceptibility to lead poisoning.Methods:A case-control study was conducted among 61 lead poisoning patients,76 lead exposed workers who had no disease of lead poisoning as in-controls and 75 people who were never exposed to lead as out-controls.The different genotypes of XRCC1 A194W and A399Q were identified by the polymerase chain reaction-restriction fragment length polymorphism(PCR-RLFP) technique.Results:The distributions of the CT+TT genotype of XRCC1 A194W polymorphism and the GA+AA genotype of XRCC1 A399Q polymorphism between lead poisoning cases and controls were significantly different(P<0.05),which showed that an increase of the susceptibility to lead poisoning was associated with the CT genotype and the TT genotype of XRCC1 A194W polymorphism(OR=2.46,95%CI=1.16~5.21).There was a decreased risk of lead poisoning for individuals carrying the GA genotype and the AA genotype of XRCC1 A399Q polymorphism(OR=0.31,95%CI=0.15~0.65).Conclusion:The gene polymorphism of XRCC1 may contribute to the susceptibility to lead poisoning.
