RT-PCR法检测外周血LUNX-mRNA诊断非小细胞肺癌微转移的研究

摘    要: 目的 探索检测中早期非小细胞肺癌外周血微转移的理想途径以及手术对促进微转移的影响。方法 对 2 0例可手术的中早期非小细胞肺癌分别于术前、手术刚结束、术后一周取外周血 ,用RT PCR法检测肺癌特异性基因LUNX的表达情况 ,并与肺部良性疾病患者外周血LUNX基因的表达进行对比。结果 在被检测的 2 0例患者中 ,有 9例在不同时期被检测到有LUNX基因表达 ,而肺部良性疾病患者外周血均未发现LUNX基因表达。结论 RT PCR法检测外周血LUNX mRNA是理想的诊断非小细胞肺癌微转移的方法 ;手术治疗可以减少肺癌患者术后远处转移的发生 ;手术操作可能促进肿瘤细胞向外周血的脱落

Abstract:Objective To set up a good method to detect the micrometastasis in peripheral blood for non small cell lung caner patients and try to find the relationship between operation and micrometastasis.Methods Totally 20 patients involved.The peripheral blood was taken before surgery, when surgery was done at once and one week after the surgery respectively. RT PCR was carried out to detect the LUNX gene expression in peripheral blood, controlled by the same method used for those patients with benign disease in the lung. Results There are 9 patients with LUNX gene expression detected by RT PCR, and none in controlled group.Conclusion RT PCR to detect the LUNX RNA in peripheral blood is an ideal method to diagnose the micrometastasis in non small cell lung cancer. Surgery may deduce the distant metastasis. The process of surgery may cause tumor cells to dismiss to peripheral blood.
