血管内皮生长因子C936T基因多态性与糖尿病肾病的关系

摘    要:目的:探讨血管内皮生长因子C936T基因多态性与中国汉族人群2型糖尿病肾病的相关性。方法:选取296例2型糖尿病患者,其中糖尿病肾病患者173例(糖尿病肾病组),2型糖尿病非肾病患者123例(糖尿病非肾病组)。采用多聚酶链反应与限制性片段长度多态性检测两组患者血管内皮生长因子C936T位点基因多态性。结果:糖尿病肾病组与糖尿病非肾病组相比,CT和TT的基因型频率显著升高(P〈0.05).并且发现T等位基因的频率在糖尿病肾病组显著高于糖尿病非肾病组(P〈0.01)。多元Logistic回归结果表明CT和TT基因型与糖尿病肾病有相关性(OR=2、532,95%CI=1.486~4.315,P=0.001)。同时目前的结果也表明CT和TT基因型对糖尿病肾病的血脂指标有影响。结论:血管内皮生长因子3'非翻译区C936T基因多态性可能作为独立危险因素在中国汉族人群糖尿病肾病的发生和发展中起重要作用。

Abstract:Objective To examine the genetic variations of the vascular endothelial growth factor(VEGF) gene and assess the possible relationship to diabetic nephropathy(DN) in type 2 diabetic mellitus (T2DM) patients in Chinese population. Methods The C936T polymorphism of VEGF gene were examined by polymerase chain reaction and restriction fragment length polymorphism in 296 Chinese subjects with T2DM,173 patients had DN and 123 were considered as control subjects (diabetic patients without DN). Results The genotype frequency of CT and TT was significantly higher in patients with DN than patients without DN (P < 0.05). And the frequency of T allele in C936T polymorphism was also found to be significantly higher in patients with DN than patients without DN (P < 0.01). Multiple logistic regression analysis revealed that TT and CT genotypes associated with diabetic nephropathy (OR=2.532, 95% CI=1.486~4.315,P=0.001). Moreover, present results suggested that CT and TT genotype had an effect on blood lipids given exposure to diabetic nephropathy. Conclusion The data suggest that the C936T polymorphism in 3’-untranslated region of VEGF could be one of the major genetic factors in increasing the risk for DN.
