参芪五味子片对阿尔茨海默小鼠的治疗作用及其对小鼠血清与海马体氧化应激水平的影响

摘    要:目的研究参芪五味子片对阿尔茨海默小鼠的治疗作用及其对小鼠血清和海马体氧化应激水平的影响。方法选取50只3月龄APP小鼠,随机平均分为模型组,阳性药组,高、中、低剂量组;另选取10只C57BL/6小鼠作为空白组。阳性药组每天灌胃给予多奈哌齐(10 mg/kg),高、中、低剂量组每天分别灌胃给予1.44、0.72、0.36 g/kg参芪五味子片,空白组和模型组则灌胃给予等量生理盐水,饲养时间为2个月。采用水迷宫实验检测各组小鼠学习与记忆能力;实验结束后取各组血清与海马体,检测其超氧化物歧化酶(SOD)、丙二醛(MDA)、总抗氧化能力(T-Aoc)与谷胱甘肽过氧化物酶(GSH-Px)水平;利用末端标记法(TUNEL法)观察小鼠海马区细胞凋亡的情况。结果高、中剂量组的潜伏期明显短于模型组,第3象限时间明显长于模型组,SOD、TAoc、GSH-Px水平明显高于模型组,MDA明显低于模型组;TUNEL染色显示,模型组与低剂量组小鼠出现较多的阳性染色的神经元,而在高、中剂量组中仅观察到少量的凋亡神经元。结论参芪五味子片对阿尔茨海默病小鼠具有较好的治疗作用,可有效改善氧化应激水平,值得进行深入研究。

Effect of shenqi wuweizi tablets in the treatment of mice with Alzheimer's diseases and its effects on the level of oxidative stress in serum and hippocampus of the mice
Abstract:Objective To study the treatment effects of shenqi wuweizi tablets in the treatment of mice with Alzheimer′s diseases and its effects on the level of oxidative stress in serum and hippocampus of the mice. Methods The 3-month APP mice (n=50) were randomly divided into model group,positive control group,high,middle and low dose group. Ten C57BL/6 mice were selected as blank control group. The mice of high,middle and low dose groups were treated with 1. 44,0. 72 and 0. 36 g/kg shenqi wuweizi tablets,while the donepezil was used in the positive control group. The mice of blank control group and model group were given the same amount of normal saline,feeding time for 2 months. Morris water maze experiment was performed to test the learning and memory ability of mice. After the ex-periment,the serum and hippocampus of each group were collected,and the levels of SOD,MDA,T-Aoc and GSH-Px were detected. The TUNEL method was used to observe the apoptosis of hippocampus of mice. Results In the water maze test,the incubation period of mice in high,middle and low dose groups were obviously shorter than that of model group. Compared with model group,the levels of SOD,T-Aoc and GSH-Px in high,middle dose groups were higher, and MDA level was lower. TUNEL staining of the mice of model and low dose group appeared more positive staining of neurons,and only a small number of apoptotic neurons were observed in high and middle dose groups. Conclusion Shenqi wuweizi tablets has good therapeutic effect on mice with Alzheimer′s disease,which can effectively improve the level of oxidative stress,and it is worth studying in depth.
