摘    要:目的:探索对脊髓腹外侧索(VLF)进行强直刺激,在运动神经元(MN)能否诱发突触传递的长时程可塑性变化。方法:应用新生大鼠脊髓切片MN细胞内记录技术,观察VLF强直刺激对在MN诱发的兴奋性突触后电位(VLF-EPSP)的影响。结果:在稳定记录到VLF-EPSP的8个MNs,给予同侧VLF强直电刺激,其中有2个MNs的VLF-EPSPs幅度增大到基础值的120%以上,且维持时间超过1 h,可以定义为长时程增强(LTP)。结论:通过强直刺激VLF,可在新生大鼠脊髓MN诱发出突触传递的长时程增强,显示了长时程突触可塑性变化。

Abstract:Objective:To investigate whether long-term plasticity of synaptic transmission could be induced in motoneuron(MN) through tetanic stimulation of ventrolateral funiculus(VLF) of spinal cord slices.Methods:The intracellular recordings were performed from MNs in neonatal rat(8-14 days old) spinal cord slices,and the electrophysiological parameters of excitatory postsynaptic potential (VLF-EPSP) in MN induced by VLF stimulation were recorded before and after tetanic stimulation of VLF.Results:After tetanic stimulation of VLF in 8 MNs,the amplitude of the VLF-EPSPs in 2 MNs increased to a maximum of 120% of the control level and both of the increase lasted for more than 1 hour,which could be referred to as long-term potentiation(LTP).Conclusion:Long-term potentiation of synaptic transmission in MN of neonatal rat spinal cord can be induced by tetanic stimulation of VLF in vitro.
