三苯氧氨和雌二醇对兔子宫内膜细胞株c-myc原癌基因表达的影响

为了解三苯氧氨(Tamoxifen)和雌二醇(E_2)对原癌基因c-myc表达的影响,在体外培养的兔子宫内膜细胞(M-60),观察到给予E_(2b)小时后,原癌基因c-myc表达明显增加。E_2浓度在10~(-10)～10~(-6)mol/L范围内,c-myc的表达随E_2浓度加大而增强。Tamoxifen可拮抗E2所引起的c-myc的表达.当Tamoxifen的浓度为10~(-5)mol/L时抑制作用最明显,Tamoxifen的最佳抑制浓度是E_2浓度的10倍。本结果揭示在体外培养系统,E_2对子宫内膜细胞的增殖作用也可能是通过c-myc基因中介的。抑制c-myc基因的表达是Tamoxifen拮抗E_2刺激子宫内膜细胞增殖的原因之一。

The influence of tamoxifen estradiol on c-myc protooncogene expression in the endometrial cell line of rabbit

The influence of tamoxifen and estradiol on expression of c-myc protooncogenes of endometrial cell line(M-60) of rabbit was studied in the present paper. Endometrial cells were incubated in a modified Eagle's medium by additional 6h incubation with estradiol or estradiol plus tamoxifen.The expression of c-myc gene was analysed by dot blotting. It was found that estradiol dose-dependently increased the expression of c-myc gene at concentrations of 10~(-10)～10~(-5)mol/L. The expression of c-myc gene induced by estradiol could be inhibited completely with 10~(-5)mol/L tamoxifen. These results suggested that the proliferation of the endometrial cell induced by estradiol in culture was mediated by c-myc gene. Inhibiting c-myc gene expression is one of the causes for tamoxifen in antagonizing the proliferative effect of estradiol on endometrial cells.
