| 摘 要： | 通过水煮提取,过AB-8树脂除杂,乙酸乙酯萃取等工艺从香椿叶中分离得到香椿叶总黄酮提取物(TFTL)。在295 nm波长下分光光度法测定TFTL体外对黄嘌呤氧化酶(XO)活性的影响,以氧嗪酸钾ip小鼠造成高尿酸血症小鼠模型,以不同剂量的TFTL连续静脉给药7天,用磷钨酸法测定TFTL对小鼠血清尿酸(SUA)水平的影响。结果表明,TFTL中黄酮含量以芦丁当量计为843.20 mg/g;TFTL在体外能够抑制XO的活性,且呈剂量依赖关系,其IC50为56.91μg/mL;在体内,中、高剂量(100、200 mg/kg)的TFTL能够极显著地降低高尿酸血症小鼠的SUA水平(P<0.001),使其恢复至正常小鼠水平(P>0.5)。
