成人骨髓基质细胞培养

为研究成人骨髓基质细胞的体外培养及其成肌特性，将来自需取髂骨的９例患者的骨髓标本进行体外培养，传代细胞进行含１０＾－８ｍｍｏｌ．Ｌ＾－１地塞米松的条件培养和不含地塞米松的非条件培养。倒置显微镜每日观察细胞生长情况，计数传代细胞中骨样细胞转化率，检测碱性磷酸酶含量。９例标本骨髓基质细胞培养均获得成功，条件培养后有较高的成骨细胞转化率碱性磷酸酶活性。结果表明，成人骨髓基质细胞条件培养后，可做为临床所需

THE CULTURE OF STROMA CELLS OF ADULT BONE MARROW

WT6BZ]In order to study the in vitro culture of adult bone marrow stroma cells and its osteogenic characteristics, nine bone marrow specimens from the ilia of the patients receiving iliac grafting were cultured in vitro, and the subcultures were cultured with conditions containing 10 8 mmol. L 1 dexamesthasone (DXM) and with no conditions. The every day growth of the cells was observed with inverted microscope, the transmission rates of the ossiform cells in the subcultured cells were counted, and the contents of AKP were measured. The bone marrow stroma cells were successfully cultured in all the nine specimens. A higher transmission rate of osteoblasts and a higher AKP activity were obtained in the culture with conditions. The results showed that the adult bone marrow stroma cells cultured with conditions may offer a source of the osteogenous seed cells for the clinical requirement.
