摘    要:目的:利用指纹图谱与药效相关性分析,研究蛇床子镇静催眠活性目标化合物。方法:采用高效液相色谱法,固定相为EliteSinoChromODS—BP5μm(4.6mm×250mm),流动相为乙腈-0.1%醋酸水溶液;梯度洗脱程序为:0~15min乙腈由23%增加到45%,15~30min乙腈由45%增加到100%,30~40min乙腈为100%;检测波长245nm(0—26min),322nm(26~30min),245nm(30~40min);柱温:40℃;建立蛇床子指纹图谱。对蛇床子采用不同极性溶剂提取,观察戊巴比妥钠持续睡眠时间,比较各提取物镇静催眠活性,并建立相应的指纹图谱。利用SPSS17.0统计分析软件计算各指纹峰峰面积与持续睡眠时间的Pearson相关系数,寻找与镇静催眠活性相关性大的指纹峰。结果:蛇床子各提取物指纹图谱中有8个共有峰,其中6#峰为欧前胡素,7#峰为蛇床子素。分析各指纹峰峰面积与镇静催眠活性相关性发现,5#峰与镇静催眠活性有显著相关性。结论:除已知具有催眠活性的蛇床子素、欧前胡素外,还发现1种镇静催眠活性目标化合物。

Abstract:Objective :To preliminarily study the material basis of sedation and hypnosis in fructus cnidii with Fingerprint and analyzing the dependablity of pharmacodynamic action, and search the active target compound. Method:The fructus cnidii fingerprints was established in high performance liquid chromatography with Elite Sino- Chrom ODS-BP 5μm(4. 6 mm × 250 mm). Mobile phase was composed of acetonitrile and 0. 1% acetic acid so- lution, gradient elution program was that acetonitrile was from 23 % to 45 % during 0 ~ 15 min, from 45 % to 100% during 15 -30 min,and 100% during 30 ~ 40min. The detection wavelength was 0 -26 min (245 nm),26 ~ 30 min (322 nm) ,and 30 ~40 min (245 nm). And the column temperature was 40 ℃ .fructus cnidii was extracted by different polar solvent, and the prolonged sleep time cooperating napental was observed. Then the activities of different extractives were compared with, and corresponding fingerprints were established. At last with statistical analysis software SPSS 17.0 Pearson correlation coefficient of the fingerprint Peaks area and the continuous sleep time was calculated to find out the fingerprint Peak which had the most correlated sedative-hypnotic activity. Re- suit:There were 8 peaks in fructus cnidii fingerprints. The 6# peak was ammidin, and the 7# peak was osthole. The 5# peak was corelated to sedative-hypnotic activity obviously. Conclusion :Anther target compound was found which had sedative-hypnotic activity as well as osthol and ammidin.
