复方川贝颗粒对OVA介导的豚鼠过敏性哮喘模型的平喘作用机制研究

摘    要:目的:探讨复方川贝颗粒对豚鼠哮喘模型的平喘作用机制.方法:70只豚鼠,随机分为7组,每组10只,雌雄各半,即模型组,复方川贝颗粒(0.67,1.34,2.68)g·kg-1组,二陈丸组(2.325 g·kg-1),地塞米松组(9.30×10-4 g·kg-1),正常对照组;试验开始第1天,豚鼠ip10%卵白蛋白(OVA)生理盐水溶液致敏,致敏后第13天灌胃给药,每日1次,给药7d,各组均按体重ig,模型组和正常对照组给予等容积蒸馏水,15 d后用1%卵白蛋白生理盐水溶液激发5次,每日1次,建立豚鼠过敏性哮喘模型,观察复方川贝颗粒对哮喘豚鼠血清中血栓素B2(TXB2)、白三烯B4(LTB4)的含量、哮喘潜伏期及肺部病理切片嗜酸粒细胞(EOS)计数.结果:与正常对照组比较,模型组豚鼠血清中的TXB2,LTB4含量明显升高(P<0.01),EOS明显增多(P<0.01).与模型组比较,复方川贝颗粒(1.34,2.68 g·kg-1)均能降低哮喘豚鼠血清中的TXB2,LTB4含量(P <0.05,P<0.01);复方川贝颗粒(0.67,1.34,2.68 g·kg-1)剂量均能减少哮喘豚鼠小气道周围EOS总数(P<0.01),延长豚鼠哮喘潜伏期(P <0.05,P<0.01).结论:复方川贝颗粒对OVA介导的豚鼠过敏性哮喘模型的平喘作用机制与调节血清炎性介质和减少EOS浸润有关.

Abstract:Objective: To investigate the anti-asthmatic mechanism of compound Chuanbei granules (CCB)in ovalbumin-mediated allergic asthma model of guinea pig. Method: Seventy guinea pigs were randomly divided into normal control,model,erchen pill(2.325 g·kg-1),dexamethasone 9.30×10-4g·kg-1 and CCB granule 0.67,1.34,2.68 g·kg-1 groups(n=10). Each group of male and female ratio was 1: 1. On the first day of the test, the guinea pigs were sensitized by intra-peritoneal injection of 10% ovalbumin (OVA) saline solution. 13 days after sensitization, corresponding drugs were given for 7 days, the model group and normal control group given distilled water, 15 days later excitation with 1% ovalbumin was carried out.After treating with the CCB granules, latent period of asthma was recorded, the amount of eosinophils(EOS) was detected, the content of thromboxane B2 (TXB2) and leukotriene B4(LTB4) were examined. Result: Compared with normal control group, in model group guinea pig serum TXB2, LTB4 level was significantly increased(P<0.01), the increased eosinophils were seen (P<0.01). Compared with model group, the CCB granules 1.34,2.68 g·kg-1 could reduce serum TXB2, LTB4 content (P<0.05, P<0.01). The CCB granule 0.67,1.34,2.68 g·kg-1 dose reduced the total number of eosinophils around the small airways of asthma guinea pig (P<0.01); and prolong the incubation period of asthma guinea pig (P<0.05, P<0.01). Conclusion: The CCB granules could play anti-asthmatic effects. The results suggest that its functional mechanism may be related to the serum inflammatoty mediators.
