凋亡相关基因Bax,Bcl-2在大鼠急性胰腺炎中表达的研究

摘    要:目的 :探讨凋亡调控基因Bax ,Bcl- 2在胰腺组织中的表达情况与急性胰腺炎严重程度的关系。方法 ;制作大鼠急性重型胰腺炎和急性轻型胰腺炎模型。制模后 3h取材 ,查血清胰淀粉酶及应免疫组化SABC法检测胰腺组织中基因Bax ,Bcl- 2蛋白表达。结果 :急性胰腺炎时 ,基因Bax蛋白表达在胰腺腺泡细胞胞浆 ,急性轻型胰腺炎组和急性重型胰腺炎组均明显高于对照组 (P <0 .0 5 ) ,且急性轻型胰腺炎组高于急性重型胰腺炎组 (P <0 .0 5 ) ;Bcl- 2蛋白表达在胰腺导管细胞胞浆 ,对照组Bcl- 2蛋白表达弱 ,急性轻型胰腺炎组和急性重型胰腺炎组表达明显增强 ,急性重型胰腺炎组高于急性轻型胰腺炎组 ,统计学处理急性轻型胰腺炎组和急性重型胰腺炎组之间无显著性差异 ,但急性轻型胰腺炎组、急性重型胰腺炎组与对照组比较差异有显著性 (P <0 .0 5 )。结论 :Bax基因在大鼠的急性胰腺炎过程中可能参与诱导胰腺腺泡细胞凋亡 ,减轻胰腺炎症反应程度 ;而Bcl- 2基因可能参与防止导管细胞凋亡而促进其增生并且形成导管管状复合体

Abstract:Objective:To investigate expression of apoptosis-regulated genes Bax and Bcl-2 in experimental acute pancreatitis (AP) and its corelation with the severity of the disease.Methods:After 3 hours of induction of the experimental pancreatitis, the rats in each group were sacrificed for detection of serum amylase and the expression of genes Bax and Bcl-2 in pancreas by SABC immunohistochemical technique.Results:In AP, Bax gene was expressed in the cytoplasm of pancreatic acinar cells. The expression in both mild and severe type of AP was significantly higher than that in the control with P <0.05,while that of the mild type was higher than the severe ( P <0.05). The Bcl-2 gene expression occurred in the cytoplasm of pancreatic duct cells in the mild and severe type which in both type was significantly higher than that in the control ( P <0.05). Though Bcl-2 gene expression in the mild type was higher than that in the severe, the difference was not statistically significant. Conclusions: Bax gene may contribute to acinar cell apoptosis and reduce the severity of acute pancreatitis. Bcl-2 gene expression in duct cells may resist to apoptosis,and might promote the cell proliferation, thus to form tubular complexes.
