脱氢表雄酮诱导SD大鼠多囊卵巢综合征模型胰岛素抵抗的研究

摘    要:目的:研究脱氢表雄酮(DHEA)诱导SD幼年雌性大鼠多囊卵巢综合征动物模型的卵巢形态学、性激素及空腹血糖和胰岛素的变化。方法:选取23日龄SD雌性大鼠,建模组每日皮下注射DHEA6mg/100g体重,对照组每日皮下注射等体积油剂。注射20天后观察两组大鼠卵巢形态学(HE染色)、性激素(E2、T、P)及空腹血糖和胰岛素的变化。结果:建模组大鼠的体重显著低于对照组(P<0.05);去除体重的影响后,建模组大鼠卵巢的重量略大于对照组,但无显著性差异(P>0.05)。建模组卵巢各级发育期卵泡及黄体少见,卵泡多呈囊性扩张,颗粒细胞层数减少,卵泡膜细胞、间质细胞增生。建模组大鼠血清E2、T、FINS、FBG均显著高于对照组(P<0.05);HOMA指数显著高于对照组(P<0.05)。结论:DHEA可诱导SD幼年雌性大鼠出现与PCOS患者相似的卵巢病理改变及雄激素升高和胰岛素抵抗等典型内分泌变化,是研究PCOS胰岛素抵抗的理想模型。

Abstract:Objective:To investigate the morphological and hormonal changes,and fasting blood insulin and fasting blood glucose's changes in the rat model of polycystic ovarian syndrome(PCOS)induced by dehydroepiandrosterone(DHEA).Methods:23-day-old female Sprague-Dawley(SD)rats were randomly divided into two groups.The rats in the experimental group were injected daily(S.C.)with 6mg/100g weight of DHEA for up to 20 days while the rats in the control group injected with the same volume of oil at the same time.Ovarian morphological changes,sex hormone(E2、T、P)levels,fasting blood insulin and glucose were detected.Results:The body weight in experimental group were lower than that in control group(P<0.05).Considering about the influence of body weight,the ovarian weight in experimental group were seemed slightly higher than that in control group but no significant difference was showed between the two groups(P>0.05).The ovaries in experimental group showed multiple follicular cysts,and the number of theca cells and interstitial cells increased while the number of granulose cells decreased.Kinds of developing follicles and corpus lutea were almost absent.The serum level of estradiol,testosterone,fasting insulin and fast glucose concentration were significantly higher in experimental group than those in control group(P<0.05),so as the HOMA index(P<0.05).Conclusion:The rat model induced by DHEA was proved to be an ideal PCOS animal model to study the development of insulin resistance of PCOS.
