早孕期外周血及蜕膜淋巴细胞CD28/CTLA-4的表达

摘    要:目的:探讨人早孕期外周血及蜕膜淋巴细胞协同刺激分子表达水平在母.胎免疫调节中的作用。方法:以15例正常育龄妇女黄体期外周血为对照,应用流式细胞仪分析15例正常人早孕期外周血及蜕膜淋巴细胞CD28和CTLA-4的表达水平。结果:各组淋巴细胞几乎不表达表面CTLA-4分子;而表达细胞内CTLA-4分子(CTLA-4i)。蜕膜组织淋巴细胞CTLA-4i^+/CD28^+的比例显著高于早孕期及黄体期外周血。早孕期与黄体期外周CTLA-4i^+/CD28^+的百分率比较无显著性差异。结论:母胎界面局部高表达细胞内CTLA-4在母-胎免疫耐受中起重要作用。

Abstract:Objective:To probe into the role of expressions of the costimulatory molecules on peripheral blood and decidual lymphocytes of early pregnancy in materno-fetal immune regulation.Methods:Fifteen women of the early pregnancy were analyzed by direct immunofluorescence flow cytometry for expression of CD28 and CTLA-4 on lymphocytes of peripheral blood and decidua,and 15 healthy fertile women of luteal phase were recruited as control.Results:All the peripheral or decidual lymphocytes hardly expressed surface CTLA-4,but expressed intracellular CTLA-4(CTLA-4i).The expression ratios of CTLA-4i+/CD28+ on lymphocyte increased significantly in decidua compared to peripheral blood of the early pregnancy and luteal phase.However there were no significant difference in the ratio of CTLA-4i+/CD28+ on peripheral lymphocyte between the early pregnancy and luteal phase.Conclusion:The hyper-expression of intracellular CTLA-4 in the decidual lymphocyte could play an important role in materno-fetal immune regulation.
