人胎盘提取液促成纤维细胞生长的研究

摘    要:目的:从人胎盘中提取一种促成纤维细胞生长的美容产品。方法:通过对健康的新鲜胎盘的粗选、匀浆、酶消化、离心、DE-52阴离子交换柱层析,获得提取物,MTT法检测各抽提成分及不同成分混合的生物学活性。结果:柱层析后可获得三个不同成分:P1、P2、P3,P3活性明显,经混合活性不明显的P2后活性明显增强。结论:建立了从胎盘中提取活性成分的方法,以混合成分的形式作为一种美容产品,不仅可增强效果,还充分利用了资源,提高了单位产量。

Abstract:Objective To extract an aesthetic product from human placenta which promotes the growth of fibroblast. Methods After rough sizing, the hygeian fresh human placenta were homogenated and then digested by enzyme. Following centrifuge, the digested products were purified by DE-52 anion exchange column ion-exchange chromatography. MTT method was used to exam the growth promoting activity of each component and their mixture. Results Via ion-exchange chromatography, three components P1, P2 and P3 were gained. P3 had obvious role on the growth activity and its role was improved significantly after mixed with P2. Conclusion A method to extract the active component from human placenta was established. By using the mixture of extracted components as aesthetic product the effect can be improved greatly and the placenta can be utilized more fully. The output was elevated relatedly.
