摘    要:目的研究不同途径骨髓基质干细胞(BMSCs)移植对小鼠糖尿病治疗效果。方法分离骨髓,贴壁培养并纯化和扩增BMSCs。腹腔注射链脲佐菌素建立小鼠糖尿病动物模型。取2×105个第3代BMSCs,对模型鼠分别进行胰腺内多点注射、尾静脉移植以及肾被膜移植。动态观察移植后小鼠的血糖、体质量及行为的变化。结果与糖尿病对照组相比,胰腺移植组、尾静脉移植组和肾被膜移植组小鼠的糖尿病症状有所改善,血糖明显降低(F=32.58~48.35,q=10.06~14.98,P<0.05),以胰腺移植组血糖下降更为显著。结论 3种途径BMSCs移植对小鼠糖尿病均有治疗作用。

Abstract:Objective To study the therapeutic effects of different ways of BMSCs transplantation on diabetic mice.Methods Mouse BMSCs were isolated,cultured and purified.A model of diabetes in mice was created by intraperitoneal injection of streptozotocin.About 2×105 BMSCs of third generation were injected into the pancreas,caudal vein and renal capsule.Blood glucose levels,body weight,and behavior were observed after transplantation of BMSCs. Results Compared with the control group,the diabetic symptoms of those treated with BMSCs transplantation improved and blood sugar decreased(F=32.58-48.35,q=10.06-14.98,P<0.05),especially for those received BMSCs transplantation in the pancreas. Conclusion All the three ways of BMSCs transplantation were effective in the treatment of diabetes in mice.
