细胞色素P4502E1和氧化应激与大鼠非酒精脂肪性肝炎的关系

摘    要:目的观察大鼠非酒精脂肪性肝炎(nonalcoholic steatohepattiis,NASH)组织病理形态学改变,探讨其与细胞色素P4502E1的表达和氧化应激的关系。方法用高脂饮食制备非酒精脂肪性肝人鼠模型,正常组普通饲料喂养,12W末处死各组大鼠,全自动生化仪测定血清ALT、AST、血糖、胰岛素,光镜下观察肝组织病理形态学改变,测定肝组织CYP450及同工酶CYP2E1含量,TBA法和黄嘌呤氧化酶法测定肝组织MDA含量和SOD活性。结果与对照组比较,模型组大鼠血清ALT、AST、血糖、胰岛素显著升高(P〈0.05),肝组织MDA及CYP2E1含量显著增高(P〈0.05),SOD活性显著降低(P〈0.05),肝脏的脂肪变性程度和炎症活动度均显著增高(P〈0.05)。结论成功复制NASH大鼠模型,CYP2E1与非酒精性脂肪性肝炎有关,MDA和SOD在NASH脂质过氧化反应中发挥重要作用。
