“三部针刺法”对类风湿关节炎模型大鼠血清中白细胞介素-17、基质金属蛋白酶9及基质金属蛋白酶组织抑制剂-1含量影响

目的:观察类风湿关节炎(Rheumatoid Arthritis,RA)模型大鼠在"三部针刺法"的干预下,血清中白细胞介素-17(Interleukin-17,IL-17)、基质金属蛋白酶(Matrix metalloproteinase,MMP)9及基质金属蛋白酶组织抑制剂-1(Tissue Inhibitor of Matrix Metalloproteinase-1,TIMP-1)含量的变化,探讨针灸通过调控细胞因子等发挥改善RA病症的作用。方法:将SPF级雄性大鼠随机分为空白组、模型组、艾灸组及针刺组,每组10只。除空白组外于右后足跖皮内注射0.15 mL弗氏完全佐剂(Freund’s Complete Adjuvant,FCA)诱发RA大鼠模型,空白组注射0.15 mL的0.9%NaCl溶液。针刺、艾灸组干预方法选用"三部针刺法",针刺组治疗每次20 min,7 d为一个疗程,间隔1天后再行第2个疗程,艾灸组艾灸时间、疗程同针刺组。治疗结束后观察大鼠体质量、右后足跖度肿胀度的变化,酶联免疫吸附测定(Enzyme Linked Immunosorbent Assay,ELISA)检测大鼠血清中IL-17、MMP9、TIMP-1含量。结果:与空白组比较,三组大鼠血清中IL-17、MMP9、TIMP-1含量皆上升(P<0.05)。与模型组比较,两干预组干预1周、2周后,血清中IL-17、MMP9、TIMP-1含量均下降(P<0.05)。与艾灸组比较,针刺组干预1周、2周后,血清中IL-17含量与艾灸组相当,血清中MMP9、TIMP-1含量均下降更明显(P<0.05)。结论:"三部针刺法"通过影响血清中IL-17、MMP9及TIMP-1的含量,来改善RA大鼠的关节炎性反应。

The effect of three-part acupuncture on serum IL-17,MMP9 and TIMP-1 in RA rats

Objective:To observe the changes of the levels of IL-17,MMP9 and TIMP-1 in the serum of rats with rheumatoid arthritis model under the intervention of three-part acupuncture,and to explore the role of acupuncture in ameliorating symptoms of RA by regulating cytokines.Methods:The SPF male rats were randomly divided into the blank group,model group,moxibustion group and acupuncture group,10 rats in each group.Except for the blank group,0.15 mL of FCA was injected intradermally into the right hindfoot plantar to induce RA rat model.The blank group was injected 0.15 mL of 0.9%NaCl solution.The acupuncture group and the moxibustion took the three-part acupuncture.The acupuncture group was treated for 20 minutes each time,a course of treatment lasted for 7 days,and the second course of treatment was performed after an interval of 1 day.The moxibustion time and treatment courses in the moxibustion group were the same as those in the acupuncture group.Results:Compared with the blank group,the levels of IL-17,MMP9 and TIMP-1 in the serum in the other three groups were all increased(P<0.05).Compared with the model group,the levels of IL-17,MMP9 and TIMP-1 in the serum were decreased in two intervention groups in 1 week and 2 weeks of treatment(P<0.05).The content of IL-17 in serum in the acupuncture group in 1 week and 2 weeks of treatment was comparable to that in the moxibustion group,and the levels of MMP9 and TIMP-1 in serum in the acupuncture group were both decreased more significantly(P<0.05).Conclusion:The three-part acupuncture intervention can significantly improve the inflammatory response of RA rats,and the mechanism may be related to affecting IL-17,MMP9 and TIMP-1 in serum.
