氧化苦参碱与国产α1 b-和α 2b-干扰素对大鼠肝星状细胞增殖和胶原合成的抑制作用

摘    要:目的研究氧化苦参碱与国产α 1b-和α 2b-干扰素对大鼠肝星状细胞增殖和胶原合成的影响,为应用氧化苦参碱防治肝纤维化提供理论依据.方法分离、培养大鼠肝星状细胞,采用MTT法和3H-脯氨酸掺人法分别测定细胞增殖和胶原合成.结果氧化苦参碱在55ug~500ug/mL,两种α-干扰素在500U-10 000U/mL浓度范围内能呈剂量依赖性抑制星状细胞的增殖和胶原合成,提高药物浓度则形成作用平台.结论氧化苦参碱与α-干扰素在体外均具有抑制大鼠肝星状细胞增殖和胶原合成的作用,这对防治肝纤维化可能有一定的临床应用价值.

Inhibition of Oxymatrine, a Chinese herbal medicine and Chinese made interferon-αl b and α2b on proliferation and synthesis of rat hepatic stellate cells
Abstract:Aim To investigate the effect of oxymatrine,a chinese herbal medicne,and interferon-α1b andα 2b on proliferation and collagen sythesis of rat stellate cells in vitro to provide theary foundation for using oxymatrine to treat live fibrosis.Method Isolate and cultrue rat hepatic stellate cells,and the cell proliferation and collagen synthesis were tested by MTT and 3H-proline incorporation,respectively.Results The inhibition of proliferation and collagen synthesis by oxymatrine at dose of 5μg/mL,and by interferon-α 1b and interferon-α 2b at 500U-10 000U/mL were observed to be in a concentration dependent manner.Conclusions The Chinese herbal medicine,oxymatrine,has the similar inhibition potential as interfer-on-α on rat stellate cells in vitro.
