反义c-myc寡核苷酸对大鼠气道平滑肌细胞周期的阻滞作用

摘    要:目的: 观察反义c-myc寡核苷酸对大鼠气道平滑肌细胞周期的影响. 方法: 大鼠气道平滑肌细胞原代培养后,以Lipofectin将反义、正义和错配c-myc寡核苷酸导入平滑肌细胞,流式细胞技术检测细胞周期,免疫组织化学方法检测cyclin D1和cyclin E的表达. 结果: 反义c-myc寡核苷酸可明显抑制大鼠气道平滑肌细胞通过S期,正义及错配c-myc寡核苷酸对细胞周期无明显影响;反义c-myc寡核苷酸抑制了大鼠气道平滑肌细胞cyclin D1和cyclin E的表达. 结论: 反义c-myc寡核苷酸可抑制大鼠气道平滑肌细胞通过S期,抑制cyclin D1和cyclin E 的表达.

Inhibitive effect of antisense oligonucleotide for c-myc on rat airway smooth muscle cell cycle
Abstract:AIM: To observe the effects of antisense oligouncleotide for c myc on rat airway smooth muscle cell cycle. METHODS: Rat airway smooth muscle cells were primarily cultured and lipofectin was used to introduce antisense oligonucleotide, sense oligonucleotide and mismatched oligonucleotide for c myc into rat airway smooth muscle cells. Cell cycles were measured by flow cytometry and the expression of cyclin D1 and cyclin E was detected immunohistochemically. RESULTS: Antisense oligonucleotide for c myc inhibited rat airway smooth muscle cells through S phase. Sense and mismatched oligonucleotides exerted no effect on cell cycles. Antisense oligonucleotide for c myc decreased the expression of cyclin D1 and cyclin E. CONCLUSION: Antisense oligonucleotides for c myc inhibit rat airway smooth muscle cells through S phase and decrease the expression of cyclin D1 and cyclin E.
