细粒棘球绦虫66kDa抗原基因在成虫吸盘上的表达

摘    要:目的 观察细粒棘球绦虫自原头蚴向成虫的生长发育过程中 6 6kDa抗原基因在虫体吸盘上和其它部位表达水平的变化。方法 用重组抗原的抗血清对原头蚴及成虫等不同发育时期的虫体进行免疫组化分析。结果 免疫组化分析显示细粒棘球绦虫 6 6kDa抗原位于各期虫体的皮层和皮下肌层 ,在虫体的表面也发现该抗原的荧光颗粒 ;在经培养 2 0h后的原头蚴和成虫的吸盘上该抗原的含量显著高于虫体的其它部位 ,但在棘球蚴内的原头蚴吸盘上该抗原含量与虫体其它部位一致 ,处于较低的表达水平。结论 原头蚴在向成虫的生长发育过程中当顶突和吸盘翻出时 6 6kDa抗原在吸盘上开始大量产出 ,提示该蛋白质可能与吸盘的吸附功能有关 ;因此该抗原有可能是一有希望的成虫疫苗候选分子。

Abstract:Aim To observe the expression of the 66 kDa antigen gene of Echinococcus granulosus during the development from protoscolex to adult Methods Immunohistochemical assay was used in this study with an anti-recombinant protein antibody Results The 66 kDa antigen was immunolocalized in the tegument and subtegumental parenchyma of every stages studied,the protein granules were also showed on the surface of the teguments The quantity of the antigen expressed was considerably higher in the sucker regions than in other parts of the adults and protoscoleces after 20 h incubation in RPMI 1640 medium,while it was expressed at a lower level in the suckers as in the other parts of the protoscoleces freshly collected from hydatid cysts Conclusion The expression of the 66 kDa antigen increased considerably in the suckers when the protoscoleces evaginated during the development to adults,suggesting that this protein is related to the attachment function of the suckers,and it may be a good candidate for the vaccine development
