摘    要:目的:探讨肿瘤坏死因子受体相关因子1(tumor necrosis factor receptor-associated factor 1,TRAF1)在正常乳腺组织、乳腺癌组织及不同侵袭能力乳腺癌细胞系中的表达情况,并分析其与微血管密度的关系。方法:应用免疫组化方法分别检测TRAF1和CD34在70例乳腺癌组织和14例正常乳腺组织中的表达,并计数微血管密度。应用Western blot方法检测TRAF1在乳腺癌细胞系MDA-MA-231(高侵袭)和MCF-7(低侵袭)中的表达。结果:TRAF1在正常乳腺组织中不表达。其细胞浆阳性表达率在非浸润性导管癌(17.14%)中高于正常乳腺组织,但差异无统计学意义(P〉0.05)。在浸润性导管癌(40%)中明显高于正常乳腺组织和非浸润性导管癌(P〈0.05)。TRAF1在乳腺癌高侵袭细胞系中的表达量高于低侵袭细胞系,但差异无统计学意义(P〉0.05)。乳腺浸润性导管癌中,TRAF1阳性表达的微血管密度明显高于阴性表达组(P〈0.05)。结论:TRAF1在乳腺癌组织中表达明显升高,与乳腺癌侵袭性相关。在浸润性导管癌中,TRAF1表达与微血管密度相关。

Abstract:Objective: To investigate the expression of tumor necrosis factor receptor-associated factor 1(TRAF1) in normal breast,breast carcinoma tissue,and cell lines with different invasive ability and the relations between TRAF1 and microvessel density(MVD).Methods: The expression of TRAF1 and CD34 in 70 specimens of breast carcinoma and 14 specimens of normal breast tissues was detected by SP immunohistochemistry separately,and MVD was counted.The expression of TRAF1 in breast cancer cell lines,MDA-MB-231 with high invasive ability and MCF-7 with low invasive ability,was detected by Western blot.Results: TRAF1 was not expressed in normal breast tissues.The cytoplasmic positive rates of TRAF1 were higher in non-invasive ductal carcinoma(17.14%) than in normal breast tissues(P>0.05),and significantly higher in invasive ductal carcinoma(40%) than in normal breast tissues and in non-invasive ductal carcinoma(P<0.05).The protein level of TRAF1 was slightly higher in MDA-MB-231 cells than in MCF-7 cells.Bat there was significont difference(P>0.05).MVD of TRAF1 positive expression was more concentrated than negative expression in invasive ductal carcinoma(P<0.05).Conclusion: The expression of TRAF1 in breast carcinoma is suppressed,and correlated to the invasive ability of breast cancer and MVD in invasive ductal carcinoma.
