不同配比辛芍组方对PC12细胞氧糖剥夺损伤的保护作用

摘    要:目的:观察不同配比辛芍组方对PC12细胞氧糖剥夺损伤的保护作用,探讨组方的最佳配比.方法:采用连二亚硫酸钠(Na2S2O4)建立PC12细胞氧糖剥夺损伤(oxygen glucose deprivation,OGD)模型,将细胞分为空白组、模型组、阳性组(尼莫地平)、辛芍药物不同配比组(灯盏细辛与赤芍质量比分别为5∶0、4∶1、3∶2、2∶3、1∶4、0∶5),检测细胞存活率、乳酸脱氢酶(LDH)释放量、超氧化物歧化酶(SOD)活性、丙二醛(MDA)和一氧化氮(N0)含量.结果:与正常组比较,模型组的细胞存活率和SOD活性显著降低(P<0.01),细胞LDH释放量、MDA和NO水平显著升高(P<0.01);与模型组比较,辛芍2∶3配比组显著提升细胞存活率(P<0.05),同时辛芍3∶2和2∶3配比组均显著降低细胞LDH释放量以及MDA和NO水平(P<0.05),提高SOD活性(P<0.05).结论:辛芍2∶3配比对PC12细胞氧糖剥夺损伤的保护作用较好.

Abstract:Objective: To investigate the optimum proportion for the formula of Herba erigerontis and Radix paeoniae rubra through comparative investigations of the protective effects on PC12 cell against oxygen glucose deprivation (OGD)-induced injury. Methods: The OGD-induced injury model was es- tablished by adopting Na2S204. Cells were divided into control group, OGD group, positive group (ni- modipine), formula group ( proportion as 5: 0,4: 1,3 : 2,2: 3,1 : 4,0: 5 ). It was aimed to detect cell survival, lactate dehydrogenase (LDH) release, superoxide dismutase (SOD) activity, malondialde- hyde (MDA) and Nitric Oxide (NO). Results: Compared with control group, the OGD model de- creased the survival rate and the activity of SOD (P 〈0.01 ), and increased the LDH release, the lev- el of MDA and NO(P 〈0.01 ) ; Compared with OGD group, the 2:3 formula group significantly in- creased the survival rate ( P 〈 0.05 ), while the 3 : 2 and 2 : 3 formula groups were significantly de- creased the LDH release, the level of MDA and NO ( P 〈 0.05 ), increased SOD activity ( P 〈 0.05 ). Conclusions: The 2:3 formula of Herba erigerontis and Radix paeoniae rubra has significant protective effects on the PC12 cells injured by OGD.
