复肝灵颗粒对鼠免疫性肝损伤的保护作用

摘    要:目的研究复肝灵颗粒(FGL)对卡介苗(BCG)加脂多糖(LPS)诱导的小鼠免疫性肝损伤的保护作用。方法给50只小鼠用BCG联合LPS腹腔注射,建立小鼠免疫性肝损伤模型,其中40只分为4组,每组10只,分别给予FGL大(100mg/kg)、中(50mg/kg)、小(25mg/kg)剂量及联苯双酯(80mg/kg)灌胃治疗,余10只模型小鼠(模型对照组)和10只未造模小鼠(正常对照组)给予蒸馏水灌胃,每天1次,6d后检测小鼠的胸腺、肝脏、脾脏脏器指数和血清谷丙转氨酶(ALT)、谷草转氨酶(AST)活性。结果模型对照组小鼠与正常对照组相比较,胸腺指数明显减小,肝脏指数及脾脏指数显著增大,血清ALT及AST活性显著增高(P均<0.01)。FGL大、中、小剂量组小鼠血清ALT及AST活性均显著降低,大、中剂量组小鼠胸腺指数显著增大(P<0.01)。结论本试验成功的制备了BCG加LPS诱导下的小鼠免疫性肝损伤模型,FGL对这种免疫性肝损伤具有显著的保护作用。

Abstract:Objective To study the effects of Fuganling(FGL) in protecting immunity hepatic injury induced by Bacille calmette guerin(BCG) and Lipopolysaccharide(LPS) in mice.Methods A total of 50 mice were viscerally injected with BCG and LPS to establish the immunological liver injury model,then they were divided into 5 groups: 4 experimental groups were given 100mg/kg,50mg/kg,25mg/kg FGL or 80mg/kg LianBenShuangZhi through a tube feeding,and the model control group was given normal saline.10 mice without injections served as the normal control group.The indexes of the thymus,liver and spleen were determined, also the activities of alanine aminotransferase(ALT),and aspartate aminotransferase(AST) in the serum were determined.Results Compared to the normal control group,the thymus index of the model control group significantly decreased,and the liver index and the spleen index significantly increased,also the activities of ALT and AST in the serum increased in a large scale(P<0.01).However,the activities of ALT and AST in the three model groups treated with FGL were significantly decreased,and the thymus index was significantly increased in the high dose and medium dose FGL groups(P<0.01).Conclusions The mouse immunity hepatic injury model was successfully induced by BCG and LPS.FGL has a remarkably protective effect on immunity hepatic injury.
