维生素E对胃黏膜损伤模型小鼠的保护作用研究

摘    要:目的:观察维生素E(VE)对胃黏膜损伤模型小鼠的保护作用。方法:取小鼠50只随机均分为正常对照组、模型组、西咪替丁(25mg·kg-1)组和VE低、高剂量(25、50mg·kg-1)组,灌胃给予相应药物30min后,后4组灌胃给予吲哚美辛25mg·kg-1,4h后处死小鼠,检测各组小鼠胃黏膜损伤程度及血清中超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量。结果:与正常对照组比较,模型组小鼠胃黏膜损伤指数和SOD活性、MDA含量均明显升高(P<0.01);与模型组比较,西咪替丁组和VE低、高剂量组小鼠胃黏膜损伤指数和SOD活性、MDA含量均明显降低(P<0.01);与西咪替丁组比较,VE高剂量组小鼠胃黏膜损伤指数和SOD活性均明显降低(P<0.01)。结论:VE可能通过抗氧化、清除氧自由基和抑制脂质过氧化过程发挥预防胃黏膜损伤的作用,且VE高剂量(50mg·kg-1)组对胃黏膜损伤的保护作用强于西咪替丁(25mg·kg-1)组。

Abstract:OBJECTIVE:To observe protective effect of vitamin E(VE) on gastric mucosal injury model mice.METHODS:50 mice were randomly divided into normal control group,model group,cimetidine group(25 mg·kg-1) and VE low-dose and high dose groups(25,50 mg·kg-1).The latter 4 groups were given indometacin 25 mg·kg-1 intragastircally 30 min after intragastrical administration.Those mice were sacrificed 4 h later.Gastric mucosal injury,SOD actirity and MDA content were determined.RESULTS:Compared with normal control group,gastric mucosal injury index,SOD activity and MDA content of model group increased significantly(P<0.01).Compared with model group,gastric mucosal injury index,SOD activity and MDA content of cimetidine group and VE groups decreased significantly(P<0.01).Compared with cimetidine group,gastric mucosal injury index and SOD activity of VE high-dose group decreased significantly(P<0.01).CONCLUSION:VE can prevent gastric mucosal injury by antioxidation and scavenging oxygen free radicals,and protective effect of VE high-dose group(50 mg·kg-1) on gastric mucosal injury is better than cimetidine group(25 mg·kg-1).
