瑞芬太尼对家兔缺血脑组织水通道蛋白-4表达的影响

摘    要:目的观察瑞芬太尼对家兔缺血再灌注损伤脑组织水通道蛋白-4(AQP-4)表达的影响。方法健康家兔25只,随机分为假手术组、缺血模型组、瑞芬太尼组。以双侧颈总动脉阻断+控制性低血压法建立家兔全脑缺血损伤模型,再灌注1h,6h,24h末,测定脑组织含水量,观察AQP4的表达,再灌注24h末,观察脑组织超微结构变化。结果与假手术组比较,模型组家兔脑组织含水量及脑组织内AQP4表达在1h,6h和24h逐渐升高(P〈0.05);与模型组比较,瑞芬太尼组脑组织含水量与AQP-4表达明显降低(P〈0.05)。结论瑞芬太尼可能通过降低脑组织内AQP-4表达,减轻脑水肿,保护脑缺血损伤。

Abstract:Objective To study the protective and resuscitative effects of remifentanil on the expression of AQP-4 in rabbits brain. Methods Twenty-five healthy rabbits were randomly divided into three groups : the sham operation group,the cerebral ischemia group and the remifentanil group. The cerebral isehemia-reperfusion injury model was established by blocking bilateral carotid artery plus the method of controlled hypotension. The moisture capacity of the brain tissue and the expression of AQP-4 was observed at the end of 1 h,6h and 24h after reperfusion. The change of ul- trastructural organization was observed at the end of 24h after reperfusion. Results Compared with sham operation group,the moisture capacity of the brain tissue and the expression of AQP-4 in the cerebral ischemia group were increased gradually after 1 h,6h and 24h(P 〈0.05 ) ;Compared with the cerebral ischemia group, the brain water content and AQP-4 expression in the remifentanil group were obviously lower( P 〈 0. 05 ). Conclusion Remifentanil may have cerebral protective effect through downregulating AQP-4 expression in cerebral ischemie area and reducing brain edema.
