血塞通注射液对局灶性脑梗死大鼠不同恢复时点大脑皮层Syp和PSD-95蛋白表达的影响

摘    要:目的观察血塞通注射液(主要成分为三七总皂苷)在不同时间点对局灶性脑梗死大鼠皮层部位突触后致密物质(postsypapticDensity-95,PSD-95)及突触素(synaptophysin,Syp)蛋白表达的影响,探索三七总皂苷对脑缺血受损神经元重塑的作用机制。方法采用改良Longa方法制备局灶性脑梗死模型,将40只SD大鼠随机等分为假手术组、模型组、三七总皂苷小剂量组、三七总皂苷大剂量组、尼莫地平组。各组动物分别于术后3d、7d、14d、28d麻醉断头取材,行免疫组织化学检测PSD-95及gyp蛋白的表达。结果模型组PSD-95蛋白表达在术后3d及7d明显降低,14d逐渐恢复正常,到28d时表达量较同时间点假手术组增高(P〈0.01);Syp蛋白在术后模型组表达亦明显降低(P〈0.01),术后14d及28d,三七总皂苷组的表达较模型组显著升高(P〈0.01)。结论三七总皂苷可以促进局灶性脑梗死后PSD及Syp蛋白表达,从而促进神经重塑与修复。

Effect of Xuesaitong Injection on PSD - 95 and Syp Protein Expression in Cortex of Focal Cerebral Infarction Rats

Abstract:Objective To observe the effect of Xuesaitong Injection on PSD- 95 and Syp protein after Middle cerebral artery occlu- sion(MCAO)operation. Methods Healthy mature male SD rats were randomly divided into model group,normal saline group,Xue- saitong small -dose group,Xuesaitong high -dose group, positive control group (nimodipine group). Each group was executed at 3 days, 7 days,4 days and 28 days and then the brain tissue was taken out to observe the expression changes of PSD- 95 and Syp by im- munohistochemistry. Results The expression of PSD- 95 in the model group was decreased at 3 and 7 days after the MCAO opera- tion,and gradually came to the normal level at 14 days (P(0.01). Syp expression also decreased obviously in model group. The ex- pression in each treatment group was higher than that in the model group. At 14th and 28th day,the expression in Xuesaitong high- dose group was significantly increased compared with that in the model group (P(0.01). Conclusion Xuesaitong Injection could in- crease the expression of PSD - 95 and Syp protein so as to promote nerve regeneration.
