CXCL12基因多态性与肿瘤易感性关联的Meta分析

摘    要:目的运用Meta分析方法综合评价CXCL12 rs1801157基因多态性与肿瘤发病的关系。方法通过计算机检索和手工检索,收集有关CXCL12 rs1801157基因多态性与肿瘤易感性关系的文献,筛选出符合条件的文献,运用Meta分析方法对各项研究进行异质性检验后计算合并OR值及其95%可信区间,并进行敏感性分析及发表偏倚的评估。结果 15篇符合条件的文献纳入本研究,累积病例组3 225例,对照组4 001例。Meta分析合并结果显示突变等位基因A携带者相比野生等位基因G携带者肿瘤发病风险显著提高,合并值OR=1.27 95%CI=1.12~1.43,差异有统计学意义(P<0.01);基因型AA∶GG,GA∶GG和(AA+GA)∶GG合并OR值及其95%可信区间分别是1.30,95%CI=1.05~1.61、1.45,95%CI=1.22~1.72和1.43,95%CI=1.21~1.68,差异均有统计学意义。按肿瘤类型进行分层,A等位基因携带者相比G等位基因携带者显著提高乳腺癌发病(OR=1.32,95%CI=1.15~1.51,P<0.01);AA∶GG,GA∶GG和(AA+GA)∶GG合并OR值及其95%可信区间分别是1.64,95%CI=1.16~2.33、1.42,95%CI=1.18~1.70和1.44,95%CI=1.21~1.72。结论 CXCL12 rs1801157基因多态性与肿瘤易感性有关,突变等位基因A与肿瘤易感性升高有关,基因型AA或基因型GA携带者相对基因型GG携带者肿瘤发病风险提高。突变等位基因A增加乳腺癌易感性。

Abstract:Objective To analysis the relationship between polymorphism gene of CXCL12 rs1801157 and tumor susceptibility by Meta-analysis.Method Collecting relevant literatures about the association between polymorphism gene of CXCL12rs1801157 and tumor susceptibility under the same criteria by computer retrieval and manual retrieval.Carrying out heterogeneity inspection by Meta-analysis,calculation the OR value and 95% confidence interval,sensitivity and publication bias were evaluated.Results 15 literatures with 3 225 cases and 4 001 controls were included.Onset risk of mutation allele A carrier was obvious higher than wild allele gene G carrier,OR=1.27,95%CI were 1.12 to 1.43,the difference had statistical significance(P<0.01).The OR value and 95%CI of genotype of AA∶ GG,GA∶ GG,OR=1.30,95%CI =1.22~1.72,(AA+GA)∶ GG,OR=1.43,95%CI=1.21~1.68.Stratified by tumor types,we found that the breast cancer incidence rate of allele A carrier was higher than allele A carrier G,OR=1.64,95%CI=1.16~2.33.GA∶ GG,OR=1.42,95%CI=1.18~1.70.AA+GA∶ GG,OR=1.44,95%CI=1.21~1.72.Conclusions CXCL12rs1801157 polymorphism gene was association with tumor susceptibility.Carriers of genotype AA or GA may association with the high risk of tumor compared with GG genetype.Allele A was a risk factor of breast cancer.
