不同产地黄芪药材中毛蕊异黄酮葡萄糖苷的定量分析

摘    要:目的通过定量分析比较不同产地黄芪药材中毛蕊异黄酮葡萄糖苷的含量,为黄芪药材的质量评价提供依据。方法采用HPLC法测定毛蕊异黄酮葡萄糖苷的含量。色谱条件:Diamonsil C18(250mm×4.6 mm,5μm)色谱柱;流动相:乙腈-0.2%甲酸水溶液,梯度洗脱;流速:1.0 mL·min-1;检测波长:260 nm;柱温:30℃;进样量:20μL。结果毛蕊异黄酮葡萄糖苷的浓度在9.92μg·mL-1~248.00μg·mL-1范围内与峰面积呈现良好的线性关系(r=0.9995);平均加样回收率为98.62%,RSD为1.89%。结论不同产地黄芪药材毛蕊异黄酮葡萄糖苷的含量存在明显差异,其中河北唐山产黄芪含量最高。利用HPLC法对指标性成分毛蕊异黄酮葡萄糖苷进行定量分析,快捷准确,可广泛用于黄芪药材的质量评价。

Abstract:Objective To determine Calycosin-7-Glucoside in Astragali Radix from different place by HPLC so as to provide experimental basis for quality evaluation of Astragali Radix.Methods HPLC was used in the quantitative analysis of Calycosin-7-Glucoside.The measurement conditions were as follows: Diamonsil C18(250 mm × 4.6 mm,5 μm) column; mobile phase: acetonitrile-0.2% formic acid aqueous solution by gradient elution; fl ow rate: 1.0 mL·min-1; detection wavelength: 260 nm,column temperature: 30 ℃.Results There was a good linear relationship between the concentration of Calycosin-7-Glucoside and absorption area value in range of 9.92 μg·mL-1~ 248.00 μg·mL-1(r =0.999 5).The average recovery was 97.70% with RSD=2.64%.Conclusion There was great difference in Calycosin-7-Glucoside content of Astragali Radix from different place.HPLC method is simple and accurate,and can be widely used in the quality evaluation of Astragali Radix.
