四君子汤对荷瘤小鼠抑瘤和诱导肿瘤细胞凋亡的实验研究

摘    要:目的 :探讨四君子汤对荷瘤小鼠的抗肿瘤作用 ,阐明该方抑瘤作用的可能机制。方法 :建立荷瘤小鼠(SRS- 82瘤株 )动物模型 ,用四君子汤煎剂灌胃。连续处理 15 d后 ,剥离瘤块称重并计算抑瘤率 ,在电镜下观察肿瘤细胞形态 ,并提取肿瘤细胞 DNA做琼脂糖凝胶电泳。结果 :四君子汤组与空白对照组比较 ,瘤重显著减轻 (P<0 .0 1)。电镜观察 ,四君子汤组的肿瘤切片大部分呈现坏死细胞的改变 ,未见有大量细胞发生凋亡特征性改变。琼脂糖凝胶电泳 ,四君子汤组电泳带未见到细胞发生凋亡的特征性 DNA梯状条带。结论 :四君子汤可以显著抑制荷瘤小鼠肿瘤生长 ,证明有明显的抑瘤作用。该方在体内没有诱导肿瘤细胞凋亡的作用 ,可能通过间接作用而发挥治疗肿瘤的效果

Abstract:Objective:To study the anti-tumor effect of sijunzi decoction on expermental tumor mice,and to clarify the mechanisms of the anti-tumor effect of sijunzi decoction.Methods:In vivo experiment,established mice bearing neoplasm(SRS-82 cell line)as the experimental tumor model,the mice were fed with sijunzi decoction.After 15 treatment days,the mice were killed.The neoplasm block was stripped and weighed,and the inhibition rate of the anti-tumor was calculated. We use electronic microscope to observe the neoplasm cells, and extract the DNA of the cells for agarose gel electrophoresis.Results:The weights of the neoplasm of sijunzi decoction group are significant lighter than those the control groups (P<0.01). But we do not find that the tumor cells of sijunzi decoction group appear special alteration which infer apoptosis. Conclusion: Sijunzi decoction has obvious anti-tumor effect in experimental tumor model.Sijunzi decoction has no effect to induce the tumor cells apoptosis.
