扎鲁司特对哮喘模型肺组织金属蛋白酶抑制剂1表达的影响

目的 观察白三烯受体拮抗剂扎鲁司特对肺组织组织金属蛋白酶抑制剂1（TIMP-1）表达水平的影响。方法 108只健康雄性豚鼠随机分为3组：哮喘发作组（A组），哮喘发作＋扎鲁司特治疗组（B组），生理盐水对照组（C）组，每组36只。每组均分为24h和1、2、4、8、12周6个小组。免疫组化标记分析肺组织中TIMP-1表达水平。结果 C组肺组织中TIMP-1的表达无明显改变。卵蛋白激发后，A组TIMP-1的表达C组稍增高，但各个时期无明显差异。B组随时间的推移，TIMP-1的表达逐渐增加，8周起B组TIMP-1表达较A组、C组明显增多，激发后12周3组之间两两比较有显著性差异（P〈0．05）。结论 扎鲁司特可以增加哮喘豚鼠肺组织TIMP-1的表达水平，白三烯受体拮抗剂对预防和减轻哮喘气道重塑有积极意义。

Effect of zafirlukast on the expression of tissue inhibitor metalloproteinase-1 in lung tissue of asthmatic guinea pig

Objective To study the effect of zafirlukast,a leukotrience receptor antagonist on the expression of tissue inhibitor metalloproteinase-1(TIMP-1) in the lungs of asthmatic guinea pig model.Methods One hundred and eight pathogen-free male guinea pigs were randomly divided into three groups:asthmatic group(A),asthmatic and zafirlukast therapeutic group(B) and normal saline control group (C).Each group was divided into six subgroups:24 hour,1 week,2 weeks,4 weeks,8 weeks and 12 weeks.The expression level of TIMP-1 in the lungs was analyzed with immunohistochemical label.Results The expression level of TIMP-1 was not significantly changed in group C.TIMP-1 expression was slightly higher in group A than that in group C after ovalbumin stimulation.As time went on,the expression level of TIMP-1 in the lungs in group B raised gradually. Eight weeks after ovalbumin stimulation,the expression level of TIMP-1 in group B was significantly higher than that in group A and C(P<0.05). Conclusion Zafirlukast increases the expression level of TIMP-1 in the lungs of asthmatic guinea pig,which may play an important role in preventing and reducing the airway remodeling.
