肾衰养真胶囊对5/6肾切除慢性肾衰竭营养不良大鼠下丘脑神经肽Y mRNA表达的影响

摘    要:目的:观察肾衰养真胶囊对5/6肾切除慢性肾衰竭(CRF)营养不良大鼠的改善作用和探讨其作用机制。方法:5/6肾切除同时予4%酪蛋白饮食制作CRF营养不良大鼠模型,观察其营养不良发生时间,符合CRF营养不良模型的随机分为正常组、模型组、肾衰养真胶囊组和开同组。药物干预4周后检测尿素氮、肌酐、白蛋白、血红蛋白、24h尿蛋白,动态观察摄食量及体重变化,灌胃4周后检测血浆神经肽Y(NPY)和下丘脑NPY mRNA表达水平。结果:CRF大鼠在术后10周末出现营养不良,与模型组比较,肾衰养真胶囊组大鼠摄食量、体重显著增高,白蛋白和血红蛋白显著升高,肾功能改善,肾衰养真胶囊可上调下丘脑NPY mRNA表达,降低血浆NPY水平。结论:肾衰养真胶囊可改善CRF营养不良其机制可能是通过上调下丘脑NPY mRNA表达及降低血浆NPY从而促进CRF营养不良大鼠摄食,增加体重。

Abstract:Objective: To investigate the effect of Shenshuai Yangzhen capsule, a preparation of traditional Chinese medicine, on malnutrition rats with chronic renal failure(CRF)and to explore the mechanism. Methods: SD rats were received 5/6 nephrectomy for preparation of CRF models, and fed 4 % easein at the same time. The blood parameters, such as serum albumin (Alh) ,urea nitrogen(BUN), serum creatinine (Scr)and haemoglobin(Hb)as well as 24 hour urineprotein (24 h Upr) , food intake and body weight in CRF rats were determined. Malnutrition initiating time was observed. Those modle rats were randomized into CRF group, kaitong group, Shenshuai Yangzhen group and normal control group. After 4 - week treatment as indicated, neusopeptide Y(NPY)mRNA level in the hypothalamus were measured by RT - PCR in rats with surgically induced renal failure (two - stage subetotal nephrectomy). The bload NPY in CRF rats were determined by radioirnmunoassay. Results: Malnutrition in CRF rats began at the end of 10 - week. Compared with those in the CRF group, the plasma NPY concentrations in Shenshuai Yangzhen group were significantly lower with substantially elevated NPYrnRNA expression in the hypothalamus and food intake. Conclusion: Shenshuai Yangzhen capsule can improve malnutrition in rats with renal insufficiency possibly by up - regulation NPYrnRNA expression in the hypothalamus and reduce the level of blood NPY.
