白藜芦醇对重症急性胰腺炎大鼠腹腔巨噬细胞功能的影响

摘    要:目的:探讨腹腔巨噬细胞(PM)在大鼠重症急性胰腺炎(SAP)中的作用,并观察白藜芦醇(Res)对SAP大鼠PM功能的影响。方法:36只大鼠随机分为假手术(SO)组、SAP组和Res组3组。制模后4、12h剖杀大鼠。分离PM并培养24h。RT—PCR法检测PM中肿瘤坏死因子-α(TNF-α)mRNA和白介素-1β(IL-1β)mRNA的表达;酶联免疫吸附法(EUSA)检测细胞培养液及血清中TNF-α和IL-1β的水平。取胰腺组织行HE染色及病理学评分。结果:与SAP组比较,Res组胰腺损伤程度明显减轻;SAP组PM中TNF—mnRNA和IL—1βmRNA的表达、细胞培养液及血清中TNF-α和IL-1β水平均高于SO组(P〈0.001);与SAP组比较,Res组PM中TNF-αmRNA和1L-1βLRNA的表达、细胞培养液及血清中TNF-α和IL-1β水平明显下降(P〈0.001),但仍高于SO组(P〈0.05)。结论:PM在SAP发病机制中起非常重要的作用;Res能显著减轻SAP时胰腺组织损伤,其机制与抑制PM功能,从而抑制TNF-α和IL-1β的分泌有关。

Effects of Resveratrol on Function of Peritoneal Macrophages in Severe Acute Pancreatitis

Abstract:Objective:To investigate the role of peritoneal macrophages (PM) in the pathogenesis of severe acute pancreatitis (SAP) in rats, and observe the effect of Resveratrol (Res) on function of PM in SAP.Methods:Thirtysix rats were divided into three groups: sham operation group(SO) ,SAP group and Res group, rats were sacrificed 4 h and 12 h after the flasking. PM were extracted, and incubated for 24 h in a humidified 5% carbon dioxide incubator. The expressions of tumor necrosis factor alpha (TNF- α) mRNA and interlealdn- 1β (IL- 1β) mRNA in PM were measured by semi - quantitative RT- PCR. The levels of TNT - α and IL - 1β in culture medium and serum were evaluated. The histological changes in pancreas were examined. Results: To contrast with SAP group, scores of pancreatic are lightened in Res group, The expressions of TNF - α mRNA and IL- 1β mRNA in PM were significantly higher in SAP group than SO group at each time point; The concentrations of TNF - α and IL- 1β in culture medium and serum were significandy increased in SAP group when compared with those in SO group ( P 〈 0.001 ). To contrast with SAP group, The expressions of TNF- αmRNA.IL- 1β mRNA in PM and the concentrations of TNF- α and IL- 1β in culture medium and serum were significantly descended in Res group ( P 〈 0.001), but all of index were higher than those in SO group ( P 〈 0.05). Conclusions: PM could play an important role in SAP pathogenesis. Res can relieve the injury of pancreas in rats with SAP, the mechanism of which may be related to the inhibitory effect of Res on the function of PM by inhibition of expression of TNF - α and IL - 1β mRNA.
