糖肽类抗生素奥利万星及其中间体生物合成研究进展

摘    要:随着抗生素耐药菌出现,新型抗生素的需求日益增加。奥利万星是近年来新上市的新型半合成糖肽类抗生素,用于敏感革兰阳性菌,包括(methicillin-resistant Staphylococcus aureus, MRSA)导致的急性细菌性皮肤和皮肤结构感染(ABSSSIs)的治疗,其优点为对耐药菌有效、半衰期长、给药方便等。本文主要对奥利万星的抗菌活性,奥利万星中间体的生物合成途径、化学结构、菌株选育等最新研究进展进行综述,为高产菌株的基因工程改造及产品开发提供指导。

Abstract:With the emergence of antibiotic-resistant bacteria, the demand for new antibiotics is gradually increasing. Oritavancin is a new type of semi-synthetic glycopeptide antibiotic developed in recent years for the treatment of acute bacterial skin and skin structure infections (ABSSSIs) caused by sensitive Gram-positive bacteria (including methicillin-resistant Staphylococcus aureus, MRSA). Oritavancin have the main advantages of being effective against drug-resistant bacteria, long half-life period, and convenient administration. In this paper, the recent research progresses, including the antibacterial activity of oritavancin, biosynthetic pathway, chemical structure, and strain screening of oritavancin intermediates, were reviewed, and will provide guidance for the genetic engineering and product development of high-yield strains.
