熊果酸抑制胃癌细胞BGC-823增殖并诱导凋亡

摘    要:目的:探讨熊果酸对人胃癌细胞BGC823的增殖抑制和诱导凋亡作用及其机制。方法:运用MTT法检测细胞的增殖;Hoechest33258荧光染色观察DNA片段化;流式细胞术分析细胞周期和凋亡率;Westernblot检测BGC823细胞内细胞色素C(cytochromeC,cytC)、survivin、caspase3的表达。结果:熊果酸对BGC823细胞具有抑制增殖和诱导凋亡作用,并呈浓度和时间依赖性,作用24h的IC50为43.10μmol·L-1;在熊果酸作用下BGC823细胞呈凋亡征象;G1期细胞数减少,细胞主要阻滞在S期;同时cytC释放和caspase3酶原活化增加,survivin表达降低。结论:熊果酸能够在体外抑制人胃癌细胞BGC823增殖并诱导其凋亡,而促进cytC释放增加,下调survivin表达,继而引起caspase3的活化可能是其作用机制之一。

Abstract:Objective: To investigate whether ursolic acid can inhibit proliferation of human gastric carcinoma cell line BGC-823 and induce apoptosis and its mechanism. Methods: Cell proliferation was determined by MTT assay. Cell apoptosis changes were observed by Hoechst 33258 fluorescent staining. Cell cycle and apoptosis rate were analysed by flow cytometry. The expression of cyt-C, survivin and caspase-3 were detected by Western blotting. Results: MTT assay showed that UA had a moderate anti-proliferation effect on BGC-823 cells in a time-and-dose-dependant manner and the IC 50 is 43.10 μmol·L -1 after 24 hour. Apparent morphological index of apoptosis was observed, and G 1 phase decline, cells mainly arrested in S phase. The expression of survivin was down-regulated, but the release of cyt-c and activation of caspase-3 were enhanced in vitro. Conclusion: Ursolicacid could inhibit proliferation and induce apoptosis in BGC-823. Down-regulation of the expression of survivin, induction of delivery of cyt-c and activation of caspase-3 may contribute to its effect.
