参附芎泽注射液对压力超负荷大鼠血流动力学和心肌胶原纤维重塑的影响研究

摘    要:目的:研究参附芎泽注射液对压力超负荷大鼠血流动力学和心肌重塑的影响。方法:复制腹主动脉缩窄大鼠模型;左心室插管术测定血流动力学指标;检测左心室质量指数(LVMI);形态学观察,HE染色光镜观察心肌结构,SABC免疫组化方法测定心肌Ⅰ型胶原(Collagen Ⅰ)和Ⅲ型胶原(Collagen Ⅲ),均摄片对比。结果:与假手术组比较,模型组左室内压力峰值(LVSP)、左心室内压最大下降速率(-LVdp/dtmax)及左心室内压最大上升速率(+LVdp/dtmax)下降;收缩压(SBP)、舒张压(DBP)、左室舒张末期压(LVEDP)均升高;LVMI增大;肌纤维增粗,CollagenⅠ和CollagenⅢ显著增加。参附芎泽注射液组与模型组比较,LVSP、±LVdp/dtmax均升高,LVEDP、SBP、DBP降低;LVMI减小;CollagenⅠ和CollagenⅢ显著减少,均有显著性差异(P<0.05或P<0.01)。结论:参附芎泽注射液对压力超负荷大鼠血流动力学和心肌胶原纤维重塑均有一定的改善作用,能预防、延缓心肌重塑的发生,提高心功能。

Abstract:OBJECTIVE: To explore the effects of Shenfu Xiongze Injection on hemodynamics and myocardial collagen remodeling in experimental pressureoverloaded rats. METHODS: Abdominal aorta coarctation model was duplicated in rats. The hemodynamics indexes were measured by introduction of a tube into rats' left ventricles; LVMI were measured. Morphology of cardiac muscle was studied and the cardiac structure was observed by HE staining and light microscopy. Collagen Ⅰ and Collagen Ⅲ were investigated by means of SABC immunohistochemial staining. Photographs of all were taken for comparison. RESULTS: In model groups as compared with the sham-operation group, LVSP, -LVdp/ dtmax and + LVdp/ d tmax decreased while SBP, DBP, LVEDP, LVMI increased, muscle fiber thickened, Collagen Ⅰ and Collagen Ⅲ increased significantly (P〈0.05) . In Shenfu Xiongze Injection group compared with the model group, LVSP and + LVdp/ dtmax increased, LVEDP, SBP and DBP decreased, LVMI decreased, Collagen Ⅰ and Collagen Ⅲ decreased significantly, all indexes showing significant differences between the two groups (P 〈 0.05 or P 〈 0.01). CONCLUSION: The present study suggested that Shenfu Xiongze Injection could improve significantly hemodynamics and myocardial collagen remodeling, prevent and retard myocardial collagen remodeling and improve cardiac function.
