摘    要:目的:观察超排卵对小鼠着床的影响和中药帮得孕的改善作用。方法:仿照临床超排卵方案,运用腹腔注射孕马血清促性腺激素(PMSG)10IU,于48h后注射绒毛膜促性腺激素(HCG)10IU的方法造模。观察妊娠第8天正常组、超排卵组、不同剂量帮得孕组小鼠胚泡着床率和平均胚泡着床点数。伊文思蓝染色观察妊娠第5天子宫内膜血管通透性情况,HE染色观察妊娠第5天子宫内膜蜕膜化情况,免疫组化和Western blot方法观察妊娠第5天子宫内膜基质金属蛋白酶-9(MMP-9)的表达。结果:与正常组相比,超排卵组着床率明显下降(P<0.05),与超排卵组相比,帮得孕各剂量组着床率均明显提高(P<0.05);超排卵抑制小鼠子宫内膜血管渗透性,延缓子宫内膜蜕膜化,抑制子宫内膜MMP-9表达,中药帮得孕则有利于促进子宫内膜血管渗透性、促进内膜蜕膜化、促进MMP-9表达。结论:超排卵可致着床障碍,与超排卵药物抑制血管渗透性、抑制子宫内膜蜕膜化和抑制内膜基质降解有关,帮得孕可以一定程度上降低和抵消超排卵这种不良的影响,有利于胚泡成功着床。

Abstract:Objective: To observe the effect of controlled ovarian hyperstimulation(COH) on mice embryo implantation and the regulatory effect of Bangdeyun,a traditional Chinese medical herbs.Methods: Following the clinical method,mice were injected with 10 IU HCG 48 h after intraperitoneal injection of 10 IU PMSG,the implantation rate and average number of implanted blastocysts were compared among normal pregnancy group,controlled ovarian hyperstimulation group,and Chinese Herbs treated group on pregnancy day 8.The vasopermeability,deciduation and matrix metalloproteinase-9(MMP-9) expression in endometrium were observed under microscope by Evans blue dying,HE stain,and immunohistochemical method respectively on pregnancy day 5.Results: Compared with that respectively in normal group,COH reduced the implantation rate obviously(P<0.05),but Bangdeyun improved implantation rate greatly(P<0.05).COH inhibited the vasopermeability of endometrium,delayed the endometrium deciduation and inhibited the expression of MMP9,but Bangdeyun can improve the vasopermeability of endometrium,endometrium decidualization and expression of MMP-9.Conclusion: Controlled ovarian hyperstimulation could cause embryo implantation dysfunction through inhibiting the vasopermeability of endometrium,delaying the endometrium decidualization,and inhibiting matix dissolving,but Bangdeyun can improve these changes and help in implantation.
