摘    要:目的探讨雷公藤甲素(TP)对2型糖尿病(T2DM)大鼠肾脏nephrin、podocin表达的影响及对TP的调节作用。方法高脂高糖喂养联合小剂量链脲佐菌素(STZ,30mg/kg)建立T2DM大鼠模型,将模型大鼠随机分为两组:模型对照组(DM组)与TP治疗组(DT组),另设正常对照组(NC组)。8周末测血糖及尿清蛋白(UAL);光镜观察肾脏病理改变,电镜观察足细胞超微结构的变化;免疫组化技术测定各组肾组织nephrin、podocin蛋白的表达。结果 DM组较NC组UAL明显升高,DT组较DM组UAL明显降低,差异均有统计学意义(P<0.01);TP可改善其病理改变及足细胞超微结构;DM组较NC组肾组织nephrin和podocin蛋白质表达均显著减少,差异有统计学意义(P<0.01),TP干预8周后可明显恢复nephrin和podocin蛋白质表达,差异均有统计学意义(P<0.01)。结论 TP可能通过恢复糖尿病大鼠肾组织nephrin与podocin蛋白表达来减少UAL排泄。

The Effect and Significance of Triptolide on the Expression of Nephrin,Podocin in Type 2 Diabetic Rat Kidneys

Abstract:Objective To observe the expression of nephfin, podocin in the kidney in type 2 diabetic rats and the effect of triptolide on it. Methods Wistar rats were fed a high fat high sugar diet for 8 weeks and given a low dose of streptozoto- tin (STZ, 30mg/kg) developed type 2 diabetic model rats. The successfully induced rats were randomly divided into two groups: type 2 diabetic mellitus without treatment group (DM) and type 2 diabetic group treated with triptolide (DT). Normal wistar rats of the same sibling were chosen as control group (NC). Renal histomorphology was observed by optic microscope and transmission electron microscope, and urinary albumin (UAL) was determined after 8 weeks. The expression of nephrin and podocin protein was examined by immunohistochemistry in the kidney. Results Increased UAL was significantly attenuated by TP treatment. Compared with those in control group, kidney protein expression of nephrin and podocin were significantly de- creased in diabetic rats, which were significantly up - regulated by tfiptolide. Conclusion Triptolide could decrease UAL in di- abetic rats, whose mechanism may be at least partly correlated with up -regulating the expression of nephrin and podocin in the kidney.
