Abstract:Objective To investigate the effects of amiodarone on testicular weight, testicular morphology, testicularenzyme activity and sperm motility in rats. Methods SD rats were randomly allocated into control group, low-dosegroup and high-dose group and continuously gavaged with amiodarone for 4 weeks. When the rats were sacrificed, thetestes of the rats were quickly separated and weighed ;testicular morphology and sperm mobility were observed bylight microscopy ; lactate dehydrogenase (LDH), succinate dehydrogenase (SDH) and nitric oxide synthase (NOS)activities and nitric oxide (NO) levels were determined by LDH, SDH, NOS and NO assay kits, respectively.Results The rats in the high-dose group showed listlessness, gray hair, decreased activity and increased aggressivebehavior. Part of ektexine of the seminiferous tubule of the testes of the rats in the high dose group was folded, andthe spermatogenic epithelial cells were irregularly arranged. In terms of testicular quality, testicular LDH activityand SDH activity, there were no statistically significant differences between the low-dose group and the high-dosegroup and the control group. In terms of NOS activity, NO content and sperm motility in testicular tissue, there wereno statistically significant differences between the low-dose group and the control group, but there were statisticallysignificant differences between the high-dose group and the control group. Conclusion High-dose amiodarone causesirregular arrangement of seminiferous tubules in the testis, increase of NOS activity and NO content in the testis,decline of sperm motility, which indicates that high-dose amiodarone may have an impact on male testis and affectmale reproductive function.
