妊娠期高血压疾病患者血浆NO水平及胎盘eNOS表达变化的意义

摘    要:目的:研究妊娠期高血压疾病患者血浆一氧化氮(nitric oxide,NO)水平及胎盘内皮型一氧化氮合酶(endo-thelia nitric oxide synthase,eNOS)表达变化的意义。方法:应用硝酸还原酶法检测妊娠期高血压疾病患者及正常妊娠妇女血浆NO3-/NO2-浓度水平(反映NO的良好指标);采用RT-PCR技术检测胎盘中eNOS mRNA的含量,对其表达水平进行半定量比较;Western blot方法检测胎盘中eNOS蛋白含量,分析各组间是否存在表达水平的差异。结果:病例组血浆中NO3-/NO2-浓度分别明显低于对照组〔(40.71±13.21)μmol/L比(54.86±8.09)μmol/L,t=4.67、P<0.001〕,病例组胎盘eNOS mRNA(66.96±5.85比102.97±6.06,t=21.51、P<0.001)和蛋白(128.54±37.61比176.19±6.99,t=6.47、P<0.001)的表达明显低于对照组。并且患者血浆一氧化氮水平和胎盘eNOS mRNA及蛋白表达随疾病严重程度呈现下降趋势。结论:血浆一氧化氮水平和胎盘eNOS mRNA及蛋白表达降低与妊娠期高血压疾病的发病有关,并可能参与了疾病的病理生理过程。

Abstract:Objective:To study the changes of plasma NO and placental eNOS expression in patients with hypertensive disorder complicated pregnancy.Methods:The plasma NO3-/NO2-was detected by spectrophotometer,which was used to provide a vivo index of NO synthesis. The mRNA and preotein of eNOS in placenta in different groups were detected by RT-PCR or Western Blot respectively.Results:The level of NO3-/NO2-in patients was lower than control group (40.71±13.21μmol/L vs 54.86±8.09μmol/L,t=4.67,P<0.001).The level of eNOS mRNA (66.96±5.85 vs 102.97±6.06,t=21.51,P<0.001) and protein (128.54±37.61 vs 176.19±6.99,t=6.47,P<0.001) in control group were higher than those in patients. According to the severity of the disease,the plasma NO and placental eNOS expression in patients with hypertensive disorder complicated pregnancy take on downtrend.Conclusion:Decreased plasma NO and placental eNOS expression may be involed in the pathogenesis of hypertensive disorder complicated pregnancy and play an important role in the pathophysiology of the disease.
